[ABCC4] The protein encoded by this gene is a member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molecules across extra- and intra-cellular membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the MRP subfamily which is involved in multi-drug resistance. The specific function of this protein has not yet been determined; however, this protein may play a role in cellular detoxification as a pump for its substrate, organic anions. Alternative splicing results in multiple splice variants encoding different isoforms. [provided by RefSeq].

Expression: According to AceView, this gene is expressed at very high level, 5.2 times the average gene in this release. The sequence of this gene is defined by 575 GenBank accessions from 553 cDNA clones, some from prostate (seen 319 times), stomach (37), kidney (22), lung (18), prostate normal (12), brain (10), 2 pooled tumors (clear cell type) (6) and 76 other tissues. We annotate structural defects or features in 46 cDNA clones.
Alternative mRNA variants and regulation: The gene contains 46 distinct gt-ag introns. Transcription produces 21 different mRNAs, 18 alternatively spliced variants and 3 unspliced forms. There are 10 probable alternative promotors, 5 non overlapping alternative last exons and 5 validated alternative polyadenylation sites (see the diagram). The mRNAs appear to differ by truncation of the 5' end, truncation of the 3' end, presence or absence of 7 cassette exons, overlapping exons with different boundaries. 206 bp of this gene are antisense to spliced gene plerdoby, raising the possibility of regulated alternate expression.
Note that mRNA .hAug10 was found in vivo, although it is a predicted target of nonsense mediated mRNA decay (NMD).
Efficacy of translation may be reduced by the presence of a shorter translated product (uORF) initiating at an AUG upstream of the main open reading frame (in variant iAug10).
Function: There are 69 articles specifically referring to this gene in PubMed. Functionally, the gene has been tested for association to diseases (Carcinoma, Small Cell; Neuroblastoma), proposed to participate in pathway (ABC transporters) and processes (ion transport, transmembrane transport). Proteins are expected to have molecular functions (15-hydroxyprostaglandin dehydrogenase (NAD+) activity, ATP binding, ATPase activity, ATPase activity, coupled to transmembrane movement of substances, chloride channel activity, nucleotide binding) and to localize in various compartments (membrane fraction, plasma membrane, platelet dense granule membrane, integral to membrane).
Protein coding potential: 15 spliced and the unspliced mRNAs putatively encode good proteins, altogether 15 different isoforms (7 complete, 3 COOH complete, 5 partial), some containing domains ABC transporter transmembrane region, ABC transporter [Pfam], some transmembrane domains [Psort2]. The remaining 5 mRNA variants (3 spliced, 2 unspliced; 2 partial) appear not to encode good proteins. Finally proteins from this gene may be modulated by acetylation; phosphorylation; ubiquitination, as detailed at PhosphoSite.
