[ACADVL] The protein encoded by this gene is targeted to the inner mitochondrial membrane where it catalyzes the first step of the mitochondrial fatty acid beta-oxidation pathway. This acyl-Coenzyme A dehydrogenase is specific to long-chain and very-long-chain fatty acids. A deficiency in this gene product reduces myocardial fatty acid beta-oxidation and is associated with cardiomyopathy. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq].

Expression: According to AceView, this gene is expressed at very high level, 6.9 times the average gene in this release. The sequence of this gene is defined by 1131 GenBank accessions, some from brain (seen 34 times), lung (33), eye (28), testis (24), pancreas (23), uterus (21), prostate (20) and 187 other tissues.
Alternative mRNA variants and regulation: The gene contains 37 distinct introns (30 gt-ag, 7 gc-ag). Transcription produces 38 different mRNAs, 36 alternatively spliced variants and 2 unspliced forms. There are 6 probable alternative promotors, 6 non overlapping alternative last exons and 15 validated alternative polyadenylation sites (see the diagram). The mRNAs appear to differ by truncation of the 5' end, truncation of the 3' end, presence or absence of 27 cassette exons, overlapping exons with different boundaries, splicing versus retention of 22 introns. 1123 bp of this gene are antisense to spliced gene DLG4, 1393 to tuwer, raising the possibility of regulated alternate expression.
4 variants were isolated in vivo, despite the fact that they are predicted targets of nonsense mediated mRNA decay (NMD).
Efficacy of translation may be reduced by the presence of a shorter translated product (uORF) initiating at an AUG upstream of the main open reading frame (in variant eAug10, mAug10, uAug10, veAug10).

Functionally, the gene has been tested for association to diseases (OMIM: acyl-coa dehydrogenase, very long-chain, deficiency of; Other sources: Cardiomyopathies; Cholesterol, HDL/blood*; Chromosome Deletion; Death, Sudden; Metabolism, Inborn Errors; Muscular Diseases; Neuropsychological Tests; Syndrome; VLCAD deficiency), proposed to participate in pathways (Fatty acid metabolism, Metabolic pathways) and processes (fatty acid beta-oxidation using acyl-CoA dehydrogenase, fatty acid metabolic process, lipid metabolic process, negative regulation of fatty acid biosynthetic process, negative regulation of fatty acid oxidation, oxidation reduction, regulation of cholesterol metabolic process, temperature homeostasis). Proteins are expected to have molecular functions (FAD binding, long-chain-acyl-CoA dehydrogenase activity) and to localize in various compartments (cytoplasm, mitochondrion, membrane, mitochondrial inner membrane). A putative protein interactor has been described (ACADVL).
Protein coding potential: 33 spliced mRNAs putatively encode good proteins, altogether 34 different isoforms (18 complete, 9 COOH complete, 7 partial), some containing domains Acyl-CoA dehydrogenase, C-terminal domain, Acyl-CoA dehydrogenase, C-terminal domain, Acyl-CoA dehydrogenase, middle domain, Acyl-CoA dehydrogenase, N-terminal domain [Pfam]. The remaining 5 mRNA variants (3 spliced, 2 unspliced; 2 partial) appear not to encode good proteins.
3 isoforms are annotated using as Met a Kozak-compatible non-AUG start, thereby gaining a minimum of 62 amino acids N-terminal to the first AUG. Finally proteins from this gene may be modulated by acetylation; phosphorylation; sumoylation; ubiquitination, as detailed at PhosphoSite.
