[ZEB1] This gene encodes a zinc finger transcription factor. The encoded protein likely plays a role in transcriptional repression of interleukin 2. Mutations in this gene have been associated with posterior polymorphous corneal dystrophy-3 and late-onset Fuchs endothelial corneal dystrophy. Alternatively spliced transcript variants encoding different isoforms have been described.

RefSeq annotates 6 representative transcripts (NM included in AceView variants .c, .d and .e), but Homo sapiens cDNA sequences in GenBank, dbEST, Trace and SRA, filtered against clone rearrangements, coaligned on the genome and clustered in a minimal non-redundant way by the manually supervised AceView program, support at least 27 spliced variants.

Expression: According to AceView, this gene is expressed at high level, 3.9 times the average gene in this release. The sequence of this gene is defined by 454 GenBank accessions from 409 cDNA clones, some from brain (seen 63 times), uterus (17), pooled human melanocyte, fetal heart, andpregnant uterus (14), human skeletal muscle (12), placenta (12), lung (11), testis (11) and 93 other tissues. We annotate structural defects or features in 36 cDNA clones.
Alternative mRNA variants and regulation: The gene contains 39 distinct gt-ag introns. Transcription produces 32 different mRNAs, 27 alternatively spliced variants and 5 unspliced forms. There are 9 probable alternative promotors, 7 non overlapping alternative last exons and 7 validated alternative polyadenylation sites (see the diagram). The mRNAs appear to differ by truncation of the 5' end, truncation of the 3' end, presence or absence of 19 cassette exons, overlapping exons with different boundaries. 398 bp of this gene are antisense to spliced gene LOC220930, raising the possibility of regulated alternate expression.
Efficacy of translation may be reduced by the presence of a shorter translated product (uORF) initiating at an AUG upstream of the main open reading frame (in variant eAug10, gAug10, mAug10).
Function: There are 60 articles specifically referring to this gene in PubMed. Functionally, the gene has been tested for association to diseases (OMIM: Corneal dystrophy, Fuchs endothelial, 6; Corneal dystrophy, posterior polymorphous, 3; Other sources: Alzheimer Disease; Corneal Dystrophies, Hereditary; Epstein-Barr Virus Infections; Neoplasms) and proposed to participate in processes (cartilage development, central nervous system development, embryonic camera-type eye morphogenesis, embryonic morphogenesis, embryonic skeletal system morphogenesis, negative regulation of cell proliferation, negative regulation of epithelial cell differentiation, negative regulation of transcription from RNA polymerase II promoter, pattern specification process, positive regulation of transcription from RNA polymerase II promoter, regulation of mesenchymal cell proliferation, regulation of smooth muscle cell differentiation, regulation of T cell differentiation in the thymus, regulation of transforming growth factor beta receptor signaling pathway). Proteins are expected to have molecular functions (chromatin binding, double-stranded DNA binding, metal ion binding, sequence-specific DNA binding, transcription factor activity, transcription factor binding, zinc ion binding) and to localize in various compartments (nucleus, intracellular, transcription factor complex). Putative protein interactors have been described (CDH1, CTBP1, CTBP2, DRAP1, EP300, KAT5, SMAD1, SMAD2, SMAD3).
Protein coding potential: 17 spliced and the unspliced mRNAs putatively encode good proteins, altogether 17 different isoforms (5 complete, 12 partial), some containing Zinc finger, C2H2 type domain [Pfam], a coiled coil stretch [Psort2]. The remaining 14 mRNA variants (10 spliced, 4 unspliced; 7 partial) appear not to encode good proteins.
6 isoforms are annotated using as Met a Kozak-compatible non-AUG start, thereby gaining a minimum of 174 amino acids N-terminal to the first AUG. Finally proteins from this gene may be modulated by phosphorylation; ubiquitination, as detailed at PhosphoSite.
