This genome was very difficult to improve due to the very high repeat content which was evident also on the ... clone level. To improve the genome, we (i) sequenced P. sojae BAC End libraries and reassembled the genome using Arachne; (ii) finished scaffolds using primer walks from subclones, transposon hopping of subclones, followed by direct BAC and fosmid sequencing; (iii) removed redundant scaffolds and linked the rest using a map generated with Illumina genotyping.  more
