The worm strain used was KH1234. This strain is mutant worm. The genotype is asd-1(yb978) III; fox-1(e2643) X. Total RNAs were extracted from synchronized L1 worms by using RNeasy Midi kit (Qiagen). Poly (A)+ RNAs were repeatedly purified with FastTrack MAG (Invitrogen), fragmented in 1Â~ Fragmentation buffer (Ambion) and reverse transcribed using random hexamers and Primescript II (Takara). Second strand cDNAs were synthesized by using RNase H (Invitrogen) and DNA polymerase I (Invitrogen). Deep sequencing was performed by utilizing Genome Analyzer II (Illumina) following standard protocols.
