Library for RNA-ligase mediated 5'-RACE. polyA+ RNA was first reacted with T4 RNA ligase and an RNA adapter without CIP pre-treatment, and thus was enriched in uncapped mRNAs. The pool was then reverse transcribed with an oligodT primer to make 1st strand cDNA. Various gene-specific primers were then used to find truncated cDNAs corresponding to the 3' fragments left by microRNA-mediated mRNA cleavage.
