Single cell microarray analysis of mouse primordial germ cells with Prdm14 mutation.

(Submitter supplied) Prdm14 is a critical gene for specifying mouse primordial germ cells (PGCs). The changes in expression in mouse PGCs caused by mutations of the Prdm14 gene were investigated at the single-cell level using microarray analysis.

Profiling of transcriptional targets of Jak1 and Stat1 in proliferating and differentiating myoblasts

(Submitter supplied) Styxl2, a poorly characterized pseudophosphatase, was identified as a transcriptional target of the Jak1-Stat1 pathway during myoblast differentiation in culture. Styxl2 is specifically expressed in vertebrate striated muscles. By gene knockdown in zebrafish or genetic knockout in mice, we found that Styxl2 plays an essential role in maintaining sarcomere integrity in developing muscles. To further reveal the functions of Styxl2 in adult muscles, we generated two inducible knockout mouse models: one with Styxl2 being deleted in mature myofibers to assess its role in sarcomere maintenance, and the other in adult muscle satellite cells (MuSCs) to assess its role in de novo sarcomere assembly. more...

Pro-cachectic factors link experimental and human chronic kidney disease to skeletal muscle wasting programs

(Submitter supplied) Skeletal muscle wasting is commonly associated with chronic kidney disease (CKD), resulting in increased morbidity and mortality. However, the link between kidney and muscle function remains poorly understood. Here, we took a complementary interorgan approach to investigate skeletal muscle wasting in CKD. We identified an increased production and elevated blood levels of soluble pro-cachectic factor Activin A, directly linking experimental and human CKD to skeletal muscle wasting programs. more...

Expression Data comparing hearts from a mouse lupus model NZB_W vs NZW controls

(Submitter supplied) Lupus nephritis with pronounced signs of chronic kidney disease leads to significant changes in the heart. It is not yet clear whether these changes are caused by the chronic kidney disease or by the autoimmune disease. It is also unclear which factors are responsible for the cardiovascular changes. Therefore, the gene expression of mice with lupus nephritis (NZB_W) will be compared with healthy controls (NZW).

Transcriptome analysis of Mapt-EGFP+ and Mapt-EGFP- myenteric cells

(Submitter supplied) Mapttm1(EGFP)Klt/J mice (Mapt-EGFP; The Jackson Laboratory, Bar Harbor, ME, USA; stock 004779) carry a knock-in of the EGFP coding sequence in the first exon of the microtubule-associated protein tau (Mapt) gene producing a cytoplasmic EGFP fused to the first 31 amino acids of MAPT. EGFP expression marks neurons including enteric neurons regardless of their lineage, closely patterning the expression of neuron-specific beta-tubulin III (TUBB3). more...

Vorinostat treatment of PPS-ICBR cells

(Submitter supplied) We generated immune checkpoint blockade resistant cell lines from a prostate tumor driven by Probasin-Cre Pten/P53/Smad4 loxp deletion, through serial implantation and ICB treatment. We then wanted to explore whether gene expression changes associated with resistance to immune checkpoint blockade could be reversed by treating ICB resistant cells with the pan-HDAC inhibitor Vorinostat. We used a microarray to determine gene expression changes caused by Vorinostat treatment in these cells with biological duplicates.

Maternal Exercise Prior to and During Gestation Induces Sex-Specific Alterations in the Mouse Placenta

(Submitter supplied) Examined the role of maternal voluntary wheel running commencing 10 weeks prior to gestation, and throughout pregnancy, on placental transcriptome in late gestation While exercise (EX) is beneficial during pregnancy for both mother and child, little is known about the mechanisms by which maternal (MAT EX) mediates changes in utero. We hypothesized that effects of MAT EX prior to and during gestation will be evident in transcriptomic signatures in the placenta and will be sexually dimorphic. more...

Expression data of skeletal stem cells from young mouse spines and femurs

(Submitter supplied) The spine is a remarkably specialized skeletal structure, exhibiting distinct anatomical phenotypes across different spinal regions. With this understanding in mind, we hypothesized that skeletal stem cells (mSSCs) originating from various spinal regions (cervical, thoracic, and lumbar) might display varying genotypic signatures that may dictate their skeletal tissue formation and shape. We also analyzed femur SSCs as an additional control.

HDAC inhibitors directly modulate T cell gene expression and signaling and promote development of effector-exhausted T cells in murine tumors [Array]

(Submitter supplied) Epigenetic regulation plays a crucial role in the development and progression of cancer, including the regulation of antitumor immunity. The reversible nature of epigenetic modifications offers potential therapeutic avenues for cancer treatment. In particular, Histone deacetylase (HDAC) inhibitors (HDACi) have been shown to promote antitumor T cell immunity by regulating myeloid cell types, enhancing tumor antigen presentation, and increasing expression of chemokines. more...

Whole genome microarray gene expression profiling of transgenic mouse hearts with four-fold increased BBLN contents

(Submitter supplied) The study performed whole genome microarray gene expression profiling of mouse hearts from BBLN-transgenic mice with four-fold increased cardiac BBLN (Bublin coiled coil protein) contents (Tg-BBLN-low) in comparison to non-transgenic FVB hearts. Tg-BBLN-low mice with four-fold increased cardiac BBLN expression under control of the myocardium-specific alpha-MHC promoter developed features of compensated heart failure with increasing age, which was characterized by a significantly reduced left ventricular ejection fraction as determined by echocardiography. more...

Expression data of PTEN deficient PKO cells

(Submitter supplied) Incurable metastatic castration-resistant prostate cancer (CRPC) eventually occurs after androgen deprivation treatment. It is important to understand how CRPC initiates/progress. We generated a mouse androgen-independent prostate cancer cell line (PKO) from PTEN null and Hi-Myc transgenic mice in C57BL/6 background. Here we analyzed the expression profiles of PKO cells.

Cdk4-E2F3 signals enhance skeletal muscle oxidative function and improve whole body metabolism

(Submitter supplied) Skeletal muscle insulin resistance and β-cell dysfunction are signature features of type 2 diabetes (T2D) pathogenesis. We previously demonstrated a pivotal role for the cell cycle kinase, Cdk4, in regulation of β-cell mass. Here, we demonstrate that Cdk4R/R mice, which harbor constitutively active Cdk4 kinase, are resistant to obesity and diabetes. The metabolic protection is associated with Cdk4 promoting the numbers and regeneration potential of slow/oxidative muscle fibers, improved muscle mitochondrial bioenergetics and elevated E2F3 and PGC-1α expression in muscle. more...
