Regulatory T cell-mediated resolution of lung injury: Identification of potential target genes via expression profiling

(Submitter supplied) We examined early and late gene expression changes using the IT LPS model of Acute Lung Injury (ALI). In this model, injury peaks at day 4 and is almost completely resolved by day 10 in wild type (WT) C57BL/6 mice. In contrast to the pattern in WT mice, lymphocyte-deficient Rag-1 -/- mice exhibit strikingly delayed resolution despite similar initial injury.

Intratracheal lipopolysaccharide (IT LPS)-induced lung injury effect on Rag-1 null, lymphocyte-deficient mutants

Temporal analysis of lungs from recombinase-activating gene-1-deficient (Rag-1 -/-), lymphocyte-deficient animals following acute lung injury. Resolution of acute lung injury is impaired in Rag-1 null mutants. Results provide insight into molecular mechanisms underlying resolution of lung injury.

Using ChIP-chip technology to reveal common principles of transcriptional repression in normal and cancer cells

(Submitter supplied) We compared 12 different cell populations, including embryonic stem cells before and during differentiation into embryoid bodies as well as various types of normal and tumor cells to determine if pluripotent vs. differentiated cell types use different mechanisms to establish their transcriptome. Ets2 is required for trophoblast stem cell self renewal

(Submitter supplied) The Ets2 transcription factor is essential for the development of the mouse placenta and for generating signals for embryonic mesoderm and axis formation. Using a conditional targeted Ets2 allele, we show that Ets2 is essential for trophoblast stem (TS) cells self renewal. Knock-in of Kras G12D in mouse MLP-29 cells

We exploited adeno-associated virus-mediated homologous recombination to insert the KRAS G12D allele in the genome of mouse somatic cells. (Submitter supplied) We created mice, which are deficient for Myc specifically in cardiac myocytes by crossing crossed Myc-floxed mice (Mycfl/fl) and MLC-2VCre/+ mice. Serial analysis of earlier stages of gestation revealed that Myc-deficient mice died prematurely at E13.5-14.5. Myc deficiency effect on the embryonic heart

Analysis of Myc-deficient hearts from embryonic day 13.5 transgenics. The E13.5 Myc-null embryos show normal ventricular size and structure, but decreased cardiac myocyte proliferation. Results provide insight into the molecular mechanisms underlying Myc-induced cellular proliferation.
