Expression data from NexCre:Atg7 conditional knockout mice brain

(Submitter supplied) Transcriptome analysis of NesCre:Atg7 conditional knockout mice. Autophagy plays an important role in regulating protein metabolism and tissue homeostasis. Recent studies have reported that neural stem cell-specific Atg7 knockout mice (NesCre:Atg7f/f cKO mice) exhibit neonatal lethal due to severe neurodegeneration. However, the precise mechanisms of how neuronal fate is regulated by the autophagic pathway have not been elucidated. more...

Effect of fasting and ketogenic diet on hippocampal astrocytes in vivo

(Submitter supplied) To understand the transcriptional effect of fasting and feeding a ketogenic diet on mouse CNS astrocytes, we performed translating ribosomal affinity purification (TRAP) of mRNAs immunoprecipitated from hippocampus. TRAP mice express a ribosomal epitope tag upon Cre-induced recombination that can be immunoprecipitated following activation. We measured the abundance of actively translating mRNAs from a ribosomal pull-down that came from adult astrocyte (Aldh1l1-Cre)-specific TRAP mice that were subjected to one of three dietary conditions: four weeks of normal chow diet, four weeks of ketogenic diet (high-fat, low-carbohydrate)43, or an 18-hour fast. more...

Exon and Gene Level Expression Profiling of Islets in Gaz Knockout Mice on Control and High Fat Diets

(Submitter supplied) Transcriptome analysis of RNA samples from islets To gain grater insight into the role and mechanisms behind the inhibitory G-Protein, Gaz we used global gene expression profiling. In mice, transgenic loss of B-cell Gaz is protective against high fat diets. through increased insulin secretion. This mechansism is poorly understood and here, using global transcriptomics we attempt to understand the genetic impact of Gaz knockout on control and HFD diets in male mice.

TBX1-FGF11 as a novel axis governing limb development

(Submitter supplied) Although intensively studied, the gene networks regulating limb development is still incomplete. Indeed, efforts to decipher the genetic and transcriptional control of limb formation rely mainly on transcriptome approaches obtained from embryos collected 0.5 or 1 day apart from E9.5 to E13.5. This approach identified gene networks tightly regulated during limb morphogenesis as well as novel genes possibly involved in these networks. more...

Exon Level Expression Profiling: a Novel Unbiased Transcriptome

(Submitter supplied) Transcriptome analysis of usp15 deficient mice RNA metabolism plays an important for regulating protein diversity. USP15, which is a deubiquitinating enzyme, is known to be a responsible gene for developmental disorders and neurodegenerative disorders. Although USP15 is implicated in RNA processing, the downstream targets regulated by USP15 have not been elucidated. Here, we performed exon array screening to identify the target of USP15 using Usp15 deficient mice. more...

Cortical astroglia transcriptomic dysregulation in the G93A SOD1 ALS mouse model

(Submitter supplied) Astroglia are the most abundant cell type in the central nervous system. They play essential roles in the homeostasis of the CNS. Astroglia are extremely diverse. In Amyotrophic Lateral Sclerosis (ALS) astroglia in spinal cord undergo dramatic transcriptomic dysregulation. However there is little to no data on the effects that cortical astroglia undergo during ALS pathogenesis. In this study we isolated pure cortical astroglia in end stage ALS mice. more...

Microarray expression data from the dissected MNTBs of FVB wild-type mice at P0, P1, P2, P3, P4, P6 and P14

(Submitter supplied) The medial nucleus of the trapezoid body (MNTB) contains a nearly homogeneous population of neurons, which are innervated by large glutamatergic nerve terminals called calyces of Held (CH). Key steps in maturation of CHs and MNTB neurons, including CH growth and competition, occur very quickly for most cells between postnatal days (P)2 and P6. Therefore, we characterized genome-wide changes in this system, with dense temporal sampling during the first postnatal week. more...

A Bifidobacteria-derived Tight adherence (Tad) pilus-associated protein signals colonic epithelial growth

(Submitter supplied) Development of the human gut microbiota commences at birth, with bifidobacteria being among the first colonizers of the newborn gastrointestinal tract. To date, the genetic basis of Bifidobacterium colonization, persistence and dialogue with the host remains poorly understood. We previously identified tight adherence (Tad) pili from Bifidobacterium breve UCC2003 as an essential colonisation factor using murine models. We have identified the protein that mediates the proliferation response, and demonstrate that bifidobacteria contribute to the maturation of the naïve gut in early life through the production of specific extracellular protein structures under in vivo conditions. more...
