Expression data from AGS cells with Helicobacter pylori infection

(Submitter supplied) Helicobacter pylori (H. pylori) is a human pathogen that infects almost half of the world’s population. Infection with H. pylori is frequently associated with chronic gastritis and can even lead to gastric and duodenal ulcers and gastric cancer. Although the persistent colonization of H. pylori and the development of H. pylori-associated gastritis remain poorly understood, it is believed that, in gastric mucosa, the modulated gastric epithelial cells (GECs) by H. more...

A role for miR-4734 in conditional suppression of ER stress-associated pro-inflammatory responses [starved vs control HFF]

(Submitter supplied) Prolonged metabolic stress can lead to severe pathologies. In metabolically challenged primary fibroblasts, we assigned a novel role for the poorly characterized miR-4734 in restricting ATF4 and IRE1-mediated upregulation of a set of pro-inflammatory cytokines and endoplasmic reticulum (ER) stress-associated genes. Conversely, inhibition of this miRNA augmented the expression of those genes. Mechanistically, miR-4734 was found to restrict the expression of NFKBIZ, that is required for optimal expression of the pro-inflammatory genes and whose mRNA is targeted directly by miR-4734. more...

Effect of miR-122 overexpression of mRNA distribution on the polyribosomes

(Submitter supplied) The aim of this study was to identify at genome-wide level, the effect of miR-122 on the distribution of its responsive genes on the polyribosomes. For this purpose human hepatoma cells Huh-7 were transfected for 48 hrs with LNA-based miR-122 inhibitors (122-PM) or miR-122 mimics (122-MIM). Following the treatment with cycloheximide, cells were lysed and layered on a 10-50% sucrose gradient. Following ultracetrifigation the polyribosomes were separated in 12 different fractions

Treatment with HKMTI-1-005, a dual inhibitor of EZH2 and G9a/GLP histone methyltransferases, promotes differentiation of acute myeloid leukaemialeukemia

(Submitter supplied) The canonical function of EZH2 is methylation of H3K27, although important non-canonical roles have recently been described. EZH2 mutation or deregulated expression has been conclusively demonstrated in the pathogenesis and response to treatment of acute myeloid leukemia (AML), thus making it an attractive therapeutic target. In this study, we therefore investigated whether inhibition of EZH2 might also improve the response of non-APL AML cells to ATRA-based therapy. more...

NPC stem-like cells [Affymetrix]

(Submitter supplied) Stem-like cells of nasopharyngeal cancer were enriched by 3 different methods: 1) Side population (SP) assay, 2) LMP2A over-expression and 3) spheroid culture. SP cells and LMP2A over-expressed cells were inoculated into immunocompromised mice for xenograft tumors. Transcriptomes of all 5 of these in vitro and in vivo generated NPC stem-like cells were then analyzed and compared to their counterparts by microarray. more...

The miR-497~195 cluster suppresses cell cycle regulators and in vivo leukemia growth in B-cell precursor Acute Lymphoblastic Leukemia [expression]

(Submitter supplied) Despite considerable improvements in the treatment of B-cell precursor acute lymphoblastic leukemia (BCP-ALL), relapse is still associated with poor prognosis. We previously found that the risk for early relapse can be predicted by the rapid engraftment of patient-derived blasts transplanted into NOD/SCID mice. In search for the cellular and molecular profile associated with this phenotype, we investigated the expression of microRNAs (miRNAs) in different engraftment phenotypes and patient outcomes. more...

Microarray gene-expression profiles of 139 pancreatic tumor,102 adjacent non-tumor tissue from patients with pancreatic ductal adenocarcinoma and 3 normal pancreas from donors.

(Submitter supplied) In order to identify key pathways associated with disease aggressiveness and therapeutics resistance in the most agrressive subset of PDAC, we analyzed gene expression profiling of tumor and adjacent non-tumor tissues from PDAC cases. Non-negative matrix factorization (NMF) clustering, using gene expression profile form PDAC tumors, revealed three patient subsets. A 142-gene signature specific to the subset with the worst patient survival, predicted prognosis and stratified patients with significantly different survival. more...

Expression data from human dental pulp stem cells and bone marrow mesenchymal stem cells

(Submitter supplied) To further reveal the mechanism of the senescence resistant property of mesenchymal stem cells from human, a gene expression profiling approach was used to compare the functional gene expression changes in BMMSCs and DPSCs with Affymetrix GeneChip Human Gene 1.0 ST arrays analysis. Using bioinformatic analysis, we aimed to identify changes in genes and pathways in human DPSCs and BMMSCs . Cells (BMMSCs and DPSCs) from healthy male subjects were seeded in 10 cm2 dishes and cultured until they reached 80 % confluence, and total RNA of DPSCs and BMMSCs were extracted and analyzed.
