Ruxolitinib inhibits Cyclosporine-induced proliferation of cutaneous squamous cell carcinoma

(Submitter supplied) Organ transplant recipients (OTRs) on Cyclosporine A (CSA) are prone to catastrophic cutaneous squamous cell carcinoma (SCC). Allograft-sparing, cancer-targeting systemic treatments are unavailable. We have shown increased risk for catastrophic SCC in OTRs via CSA-mediated induction of Interleukin-22 (IL-22). Herein, we found CSA drives SCC proliferation and tumor growth through IL-22 and JAK/STAT pathway induction. more...

Sequential gene regulatory events leading to glucocorticoid-evoked apoptosis of CEM human leukemic cells [HG_U95Av2]

(Submitter supplied) Glucocorticoid (GC)- sensitive cells are killed by receptor (GR)-filling concentrations of GC for a sufficient time. We used 3 closely related clones (2 sensitive, 1 resistant) from the CEM line of ALL cells to test the hypothesis that over time, a distinct set of regulated genes would be found in sensitive clones, with cross-talk to other significant pathways

Global expression changes of LNCaP and PC3 cells in response to acute Notch activation

(Submitter supplied) Adenocarcinomas of the prostate arise as multifocal heterogeneous lesions as the likely result of genetic and epigenetic alterations and deranged cell-cell communication. Notch signaling is an important form of intercellular communication with a role in growth/differentiation control and tumorigenesis. Contrasting reports exist in the literature on the role of this pathway in prostate cancer (PCa) development. more...

caArray_merco-00121: In silico dissection of cell-type-associated patterns of gene expression in prostate cancer

(Submitter supplied) Prostate tumors are complex entities composed of malignant cells mixed and interacting with nonmalignant cells. However, molecular analyses by standard gene expression profiling are limited because spatial information and nontumor cell types are lost in sample preparation. We scored 88 prostate specimens for relative content of tumor, benign hyperplastic epithelium, stroma, and dilated cystic glands. more...

caArray_louis-00379: Gene Expression-based Classification of Malignant Gliomas Correlates Better with Survival than Histological Classification

(Submitter supplied) Microarray analysis was used to determine the expression of 12,000 genes in a set of 50 gliomas, 28 glioblastomas and 22 anaplastic oligodendrogliomas. Supervised learning approaches were used to build a two-class prediction model based on a subset of 14 glioblastomas and 7 anaplastic oligodendrogliomas with classic histology. A 20-feature k-nearest neighbor model correctly classified 18 of the 21 classic cases in leave-one-out cross-validation when compared with pathological diagnoses. more...

caArray_lavar-00111: SPECIFIC GENE EXPRESSION PROFILES ASSOCIATED WITH INFANT DISSEMINATED NEUROBLASTOMA

(Submitter supplied) Neuroblastoma (NB) tumours have the highest incidence of spontaneous remission, especially among the stage 4s NB subgroup affecting infants. Clinical distinction of stage 4s from lethal stage 4 can be difficult, but critical for therapeutic decisions. The aim of this study was to investigate differential gene expression amongst infant disseminated NB subgroups. Thirty-five NB tumours from patients diagnosed at < 18 months (25 stage 4 and 10 stage 4s), were evaluated gene expression analyses. more...

caArray_lavar-00109: Differential expression of genes characterize neuroblastic tumours with distinct ploidy status

(Submitter supplied) Neuroblastic tumours (NBTs) represent a heterogeneous spectrum of neoplastic diseases associated with multiple genetic alterations. Structural and numerical chromosomal changes are frequent and are predictive parameters of NBTs outcome. We performed a comparative analysis of the biological entities constituted by NBTs with different ploidy status. Gene expression profiling of 49 diagnostic primary near-triploid (n= 22) and near-diploid/tetraploid (n=27) NBTs performed using Affymetrix U95Av2 arrays, revealed distinct expression profiles associated with each NBT subgroup. more...

caArray_golub-00142: Gene expression correlates of clinical prostate cancer behavior

(Submitter supplied) Prostate tumors are among the most heterogeneous of cancers, both histologically and clinically. Microarray expression analysis was used to determine whether global biological differences underlie common pathological features of prostate cancer and to identify genes that might anticipate the clinical behavior of this disease. While no expression correlates of age, serum prostate specific antigen (PSA), and measures of local invasion were found, a set of genes was identified that strongly correlated with the state of tumor differentiation as measured by Gleason score. more...

caArray_becic-00235: Gene expression alterations in prostate cancer predicting tumor aggression and preceding development of malignancy

(Submitter supplied) The incidence of prostate cancer is frequent, occurring in almost one-third of men older than 45 years. Only a fraction of the cases reach the stages displaying clinical significance. Despite the advances in our understanding of prostate carcinogenesis and disease progression, our knowledge of this disease is still fragmented. Identification of the genes and patterns of gene expression will provide a more cohesive picture of prostate cancer biology. more...

Keratinocyte detachment-differentiation connection

(Submitter supplied) Epidermis, a continuously self-renewing and differentiating organ, produces a protective stratum corneum that shields us from external chemical, physical and microbial threats. Epidermal differentiation is a multi-step process regulated by influences, some unknown, others insufficiently explored. Detachment of keratinocytes from the basement membrane is one such pro-differentiation stimulus. Here, we define the transcriptional changes during differentiation, especially those caused by detachment from the substratum. more...

Expression data from human abdominal, subcutaneous adipose tissue

(Submitter supplied) FCHL is a common, complex genetic lipid disorder with a largely unknown aetiology. Altered adipose tissue metabolism has been implicated as contributing to FCHL. We used microarrays to investigate the mRNA profiles of FCHL patients with CHD and non-FCHL patients with CHD in comparison with non-FCHL, non-CHD patients to define a core set of genes that were differentially expressed in FCHL and to provide a reasonable launch-pad for examining the direct contribution of one of this gene set to the development of adipose tissue.

Analyses of a deactivation genetic variation in Ha-Ras proto oncogene identified in a patient wit premature aging and insulin resistance

(Submitter supplied) Inherited genetic variants of insulin receptor induced cellular signaling have long been suspected to contribute to the development of type-2- diabetes mellitus. In this report we discuss a heterozygous mutation in the first coding exon of the proto-oncogene Ha-Ras (Ha-RasA11P) that we have identified in a patient with familial premature aging syndrome. The patient has atopic sklerodermic skin alterations, insulin resistance as well as disturbances in lipid metabolism. more...

A mutation in the c-Fos gene associated with congenital generalized lipodystrophy

(Submitter supplied) Lipodystrophies resemble syndromes of disturbed adipocyte biology or development and severe congenital forms (CGL) lack adipose tissue. The ubiquitous immediate-early gene c-fos is one essential transcription factor to initiate adipocyte differentiation. In a CGL patient we identified a single homozygous point mutation in the promoter of c-fos gene. The mutation facilitates the formation of a novel specific protein/ DNA complex and ubiquitously reduces basal and inducible c-fos transcription activity. more...
