Nerve Growth Factor neutralization promotes oligodendrogenesis by increasing miR-219a-5p levels

(Submitter supplied) Alterations of the crosstalk between the nervous and immune systems may underlie the inflammatory and neurodegenerative changes characterizing Multiple Sclerosis (MS). Among the molecules that actively interconnect these two systems is the Nerve Growth Factor (NGF), involved in both regulating inflammation and in brain homeostasis. Additionally, NGF is known to modulate oligodendrogenesis and reduce myelination, further suggesting an involvement of NGF in MS ethiology. more...

Differentially expressed microRNAs in antigen-specific Th17 cell cultures from WT and CD69KO mice

(Submitter supplied) To explore miRNA signature in T cells during moderate (wild type BALB/c) and severe (cd69-/- in BALB/c background) EAM development, we performed microarray experiments with OVA-specific Th17 (Th17 Ag-sp) cell skewed cultures. The comparison of miRNA profiles from Th17 Ag-sp subsets of cd69-/- and wild type mice, with severe and mild EAM respectively, revealed a number of deregulated miRNAs.

Characterization of microRNA signature in activated CD4+ T, polyclonal Th17 (Th17 poly), OVA-specific Th17 (Th17 Ag-sp) cell skewed cultures

(Submitter supplied) To determine specific miRNAs for Th17 cells, we performed comparative miRNA screening of activated CD4+ T, polyclonal Th17 (Th17 poly) and OVA-specific Th17 (Th17 Ag-sp) cell skewed cultures or Th17 Ag-sp sorted cells. The expression of miRNAs analyzed by microarray revealed 27 miRNAs exclusively expressed in culture or sorted Th17 Ag-sp cell subset.

MicroRNA profiling of demyelination and remyelination areas in corpus callosum of cuprizone-treated mice

(Submitter supplied) Mouse cuprizone (CPZ ) model of experimental de- and remyelination was applied to mimic demyelination pathology of multiple sclerosis. In order to identify differentially expressed microRNAs involved in de- and remyelination, the affected areas of corpus callosum were isolated from mice exposed to CPZ and conducted an Agilent microarray analysis. To induce demyelination, CPZ was administrated for four weeks. more...

MicroRNA expression profiling of prefrontal cortex samples from C57 mice treated with saline or morphine

(Submitter supplied) Our previous study showed that Dicer1 expression is positively correlated with the development of analgesic tolerance. To further understand the miRNA regulation by Dicer in the development of tolerance, we performed microRNA profiling using Agilent mouse miRNA arrays to identify the miRNA profiles in the prefrontal cortex from C57BL/6J (C57) mice under saline or morphine treatment.

Identification of a novel PPARβ/δ / miR-21-3p axis in UV-induced skin inflammation [mouse miRNA]

(Submitter supplied) Although excessive exposure to UV is widely recognized as a major factor leading to skin perturbations and cancer, the complex mechanisms underlying inflammatory skin disorders resulting from UV exposure remain incompletely characterized. The nuclear hormone receptor PPARβ/δ is known to control cutaneous repair and UV-induced cancer development. Here, we describe a novel PPARβ/δ-dependent molecular cascade involving TGFβ-1 and miR-21-3p, which is activated in the epidermis in response to UV exposure. more...

miRNA expression patterns in immature and CpG stimulated murine BMDCs

(Submitter supplied) Toll-like receptor (TLR)-induced maturation of dendritic cells (DCs) leads to the production of proinflammatory cytokines as well as the upregulation of various molecules involved in T cell activation. These are believed to be the critical events that account for the induction of the adaptive immune response. Here, we have examined the role of micro-RNA-155 (miR-155) in DC function and the induction of immunity. more...

Identification of microRNAs involved in acute rejection and spontaneous tolerance in murine hepatic allografts

(Submitter supplied) Graft acceptance without the need for immunosuppressive drugs is the ultimate goal of transplantation therapy. In murine liver transplantation, allografts are accepted across major histocompatibility antigen complex barriers without the use of immunosuppressive drugs and constitute a suitable model for research on immunological rejection and tolerance. MicroRNA (miRNA) has been known to be involved in the immunological responses. more...

Metabolic programs orchestrated by the activated Ha-ras and Îroteins in mouse liver tumors [miRNA]

(Submitter supplied) The process of hepatocarcinogenesis in the diethylnitrosamine (DEN) initiation/phenobarbital (PB) promotion mouse model involves the selective clonal outgrowth of cells harboring oncogene mutations in Ha-ras, B-raf, or Ctnnb1. Here, we have characterized mouse liver tumors harboring either Ctnnb1 or Ha-ras mutations via integrated molecular profiling at the transcriptional and translational and post-translational levels. more...

microRNA expression during ovine adult mammary gland development

(Submitter supplied) The mammary gland undergoes extensive remodeling between the begin- ning of pregnancy and lactation; this involves cellular processes including cell proliferation, differentiation, and apoptosis, all of which are under the control of numerous regulators. To unravel the role played by miRNA, we describe here 47 new ovine miRNA cloned from mammary gland in early pregnancy displaying strong similari- ties with those already identified in the cow, human, or mouse. more...

Androgen-Responsive microRNAs in mouse Sertoli Cells

(Submitter supplied) Although decades of research have established that androgen is essential for spermatogenesis, androgen’s mechanism of action remains elusive. This is in part because only a few androgen-responsive genes have been definitively identified in the testis. Here, we report that microRNAs—small, noncoding RNAs—are one class of androgen-regulated trans-acting factors in the testis. Specifically, by using androgen suppression and androgen replacement in mice, we show that androgen regulates the expression of several microRNAs in Sertoli cells. more...

LIN28A-mediated regulation of microRNA biogenesis in mouse embryonic stem cell (A3-1) [Agilent]

(Submitter supplied) LIN28A is a highly-conserved RNA-binding protein which is known to be involved in embryonic development, stem cell maintenance and proliferation. LIN28A is expressed in various types of cancer, and they are associated with advanced tumor malignancy. In embryonic stem cell, LIN28A specifically binds to let-7 precursors to suppress biogenesis of the let-7 microRNA family. In addition, LIN28A was reported to bind several mRNAs such as Oct4, cyclin A/B and histone H2A to activate their translation. more...
