Loss of Feedback Regulation between FAM3B and Androgen Receptor Driving Prostate Cancer Progression

(Submitter supplied) TMPRSS2-ERG gene fusion is one of the most frequent genetic alterations in prostate cancer (PC). Through a systematical survey of the loss of interstitial genes and patient outcomes, we nominated FAM3B as the top-ranked interstitial genes whose loss is associated with a poor prognosis. Using cultured cells, patient- and cell-line-derived xenograft models in gonad-intact or castrated host, and genetically engineered mouse models with FAM3B overexpression, knockdown, or knockout, we defined a driver role of FAM3B loss in prostate cancer progression. more...

Distinct roles of the two SEC scaffold proteins, AFF1 and AFF4, in regulating RNA Pol II transcription elongation [RNA-seq 2]

(Submitter supplied) The P-TEFb-containing super elongation complex (SEC) plays the essential role in transcriptional elongation control. The AF4/FMR2 family members AFF1 and AFF4 are the central scaffold proteins of SEC, associated with different human diseases. However, their specific roles in transcriptional control remain unclear. Here, we report that AFF1 and AFF4 show distinct genomic distribution patterns around TSS. more...

Inverse Agonists of the Androgen Receptor

(Submitter supplied) We report the efficacy of androgen receptor inverse agonists (ARIAs) in altering androgen receptor (AR) transcriptional activity. Through RNA-sequencing studies in a panel of prostate cancer cell lines and patient derived xenograft models, we demonstrate ARIAs succcessfully inhibit androgen response gene sets. ChIP-seq and CUT&RUN studies demonstrate H3K27ac decreases at AR-target genes in response to treatment. more...

Differential Expression of Calcium Channels and Signaling Pathways in Hematologic Cancers

(Submitter supplied) Cell surface calcium (Ca2+) channels permit Ca2+ ion influx, with Ca2+ taking part in cellular functions such as proliferation, survival, and activation. The expression of voltage-dependent Ca2+ (CaV) channels may modulate the growth of hematologic cancers. Profile analysis of Ca2+ channels, with a focus on the L-type CaV channels, was performed on RNA sequencing data from lymphoma and leukemia cell lines and samples derived from patients with diffuse large B cell lymphoma (DLBCL). more...

ERa-related enhancers driven by BAP18-induced chromatin accessibility with CTCF/NURF complex enrichment contributes to aromatase inhibitor non-response in breast cancer (ChIP-seq)

(Submitter supplied) Endocrine therapy is the preferred therapeutic strategy for estrogen receptor alpha (ERa)-positive breast cancer, but intrinsic endocrine resistance leads a significant challenge. CCCTC binding factor (CTCF)-mediated hyperactivation of ERa-associated enhancers have been recognized as important molecular mechanism leading to endocrine resistance, yet the underlying molecular mechanism of this process still remains unclear. more...

Targeting refractory mantle cell lymphoma for imaging and therapy using C-X-C chemokine receptor type 4 radioligands

(Submitter supplied) CXCR4 is widely expressed in hematological malignancies, including MCL The Cancer Cell Line Encyclopedia (CCLE) showed relatively high levels of CXCR4 mRNA in hematological malignancies but had no MCL cell lines within its collection. To better assess the suitability of targeting CXCR4 in MCL, we screened 11 MCL cell lines for CXCR4 mRNA expression (Cancer Research Centre, CRC). For comparison, we profiled other lymphoma subtypes, including Burkitt (n=3), activated B cell-like (ABC) diffuse large B-cell lymphoma (DLBCL) (n=6), and germinal center B cell-like (GBC) DLBCL (n=14). more...
