Improved salt tolerance of Synechococcus elongatus PCC 7942 by heterologous synthesis of compatible solute ectoine

(Submitter supplied) In order to explore the effect of introducing the ectoine synthesis module in Synechococcus elongatus PCC 7942 on the global regulation of the strain under high-salt environment. The ectoine synthesis module was introduced at the Synpcc7942_0808 locus to obtain the Syn7942/Δsps-ect strain that lacked sucrose synthesis and could synthesize ectoine. Comparative transcriptomic analysis was performed on WT and Syn7942/Δsps-ect under 0 mM and 300 mM NaCl conditions.

An RpaA-dependent sigma factor cascade sets the timing of circadian transcriptional rhythms in Synechococcus elongatus

(Submitter supplied) The circadian clock of the cyanobacterium Synechococcus elongatus PCC 7942 drives oscillations in global mRNA abundances with 24 hour periodicity under constant light conditions. Measurement of transcription factor binding in cyanobacteria grown under simluated natural light conditions [ChIP-Seq]

(Submitter supplied) We used ChIP sequencing to measure genome-wide binding of transcription factors in the cyanobacterium Synechococcus elongatus PCC 7942 grown under dynamic light regimes that mimic the rapid changes in light intensity that accompany changes in shading. Analysis of gene expression in the "clock rescue" and rpaA- "clock rescue" strains of Synechococcus elongatus PCC7942 with RNA sequencing in light/dark conditions

(Submitter supplied) The transcription factor RpaA is the master regulator of circadian transcription in cyanobacteria, driving genome-wide oscillations in mRNA abundance. Deletion of rpaA has no effect on viability in constant light conditions, but renders cells inviable in cycling conditions when light and dark periods alternate. We investigated the mechanisms underlying this viability defect. We performed RNA-seq experiment using the rpaA- “clock rescue” and “clock rescue” strains and we show that the rpaA- “clock rescue” strain is defective in transcription of genes crucial for utilization of carbohydrate stores at night.
