Critical gene filtration for the caviar yield of hybrid sturgeon

(Submitter supplied) Hybrid sturgeon, an ancient fish, has great economic and nutritional value as a source of caviar. Although significant differences exist in the female reproductive capacity under artificial breeding conditions, differential gene expression associated to domesticated female reproductive capacity have never been investigated. In this study, we conducted reference genome-based transcriptome sequencing of 16 individuals from high-yield (H) and low-yield (L) populations, respectively.

Ancient vertebrate dermal armor evolved from trunk neural crest

(Submitter supplied) Bone is an evolutionary novelty of vertebrates, likely to have first emerged as part of ancestral dermal armor that consisted of osteogenic and odontogenic components. Whether these early vertebrate structures arose from mesoderm or neural crest cells has been a matter of considerable debate. To examine the developmental origin of the bony part of the dermal armor, we have performed in vivo lineage tracing in the sterlet sturgeon, a representative of non-teleost ray-finned fish that has retained an extensive postcranial dermal skeleton. more...

Comparison of RNA localization during oogenesis within Acipenser ruthenus and Xenopus laevis [Acipenser ruthenus]

(Submitter supplied) In this study we analyzed the spatial and temporal localization of maternal transcripts during oogenesis in Acipenser ruthenus. The occurrence of transcript asymmetry in A. ruthenus has been described at a global level only in matured eggs. However not much is known about the asymmetry during oogenesis. In this study we assessed the temporal establishment of the transcript localization at a global level for A. more...

Transcriptomic analysis of sturgeon embryos in response to chemically induced DNA damage

(Submitter supplied) In this study we show that the exposure to genotoxicants (10 nM camptothecin and 20 µM Olaparib) affects embryo development in a stage-dependent manner with early stages being the most sensitive to DNA damage. The transcriptomic and proteomic patterns changed significantly after exposure to genotoxicants compared to the control. Camptothecin and Olaparib downregulated metabolic pathways and developmental processes. more...

Diverse CGG binding proteins across eukaryotes produced by independent domestications of hAT transposons

(Submitter supplied) The human transcription factor (TF) CGGBP1 (“CGG Binding Protein”) is conserved only in amniotes, and is believed to derive from the zf-BED and Hermes transposase DNA-binding domains (DBDs) of a hAT DNA transposon. Here, we examine the DNA binding preferences of a wide variety of metazoan CGGBP1-like TFs with this bipartite domain using PBM experiments. The derived motifs are available at ...

Transcriptome profiles of sturgeon lateral line electroreceptor and mechanoreceptor during regeneration

(Submitter supplied) The larvae were treated with 200 μM neomycin to deplete the hair cells in neuromast and ampullary organ. Two kinds of sensory epithelia at the ventral side of head were separated at 12 hour-post treatment (hpt) and 24 hpt, respectively. Mechano- and electro- sensory epithelia before neomycin treating were collected respectively as control (untreated). The epithelia near but without two kinds of sensors (EPs) were also collected. more...

RNA_Seq analysis of localization along animal-vegetal axis of Acipenser ruthenus

(Submitter supplied) We performed cryosectioning of oocytes along the animal-vegetal axis (first developmental axis, section A (first animal) to section E (last vegetal), followed by RNA-Seq to determine the localization profiles of coding and noncoding RNAs. The method allowed for a complete view on RNA localization. We found that nearly all RNAs are localized, but only a small percentage is actively transported during oogenesis.

Comparative study of gene expression during early vertebrate development between Xenopus laevis and Sturgeon

(Submitter supplied) Sturgeon and Xenopus is estimated to be separate by about 400 My of evolution. We have collected data on temporal expression profile in Sturgeon in order to compare the level and timing of expression for homologous pairs of genes, commonly referred to as hetero-metry and hetero-chrony. Following up our previously published research on similar comparison between two species of Xenopus separated by estimated 50 My, we are asking whether patterns of expression and signaling cascades of two key developmental periods - gastrulation and neurulation are implemented via conserved molecular basis. more...

Transcriptome profiling reveals cleft palate-related genes in Chinese sturgeon (Acipenser sinensis)

(Submitter supplied) The Chinese sturgeon (Acipenser sinensis) is anadromous fish distributed in Yangtze River and East China Sea. In this study, we reported cleft-palate Chinese sturgeons in artificial population for the first time. In order to explore the genetic basis of palate malformation in A. sinensis, Illumina RNA-seq technology was used to analyze the transcriptome data of normal and cleft-palate individuals in farmed Chinese sturgeons. more...

Insights into electrosensory organ development, physiology and evolution from a lateral line organ-enriched transcriptome

(Submitter supplied) The anamniote lateral line system, comprising mechanosensory neuromasts and electrosensory ampullary organs, is a useful model for investigating the developmental and evolutionary diversification of different organs and cell types. Zebrafish neuromast development is increasingly well understood, but neither zebrafish nor Xenopus is electroreceptive and our molecular understanding of ampullary organ development is rudimentary. more...

High conservation in transcriptomic and proteomic response of white sturgeon to equipotent concentrations of 2,3,7,8-TCDD, PCB 77, and benzo[a]pyrene

(Submitter supplied) Adverse effects associated with exposure to dioxin-like compounds (DLCs) are mediated primarily through activation of the aryl hydrocarbon receptor (AHR). However, little is known about the cascades of events that link activation of the AHR to apical adverse effects. Therefore, this study used high-throughput, next-generation molecular tools to investigate similarities and differences in whole transcriptome and whole proteome responses to equipotent concentrations of three agonists of the AHR, 2,3,7,8-TCDD, PCB 77, and benzo[a]pyrene, in livers of a non-model fish, the white sturgeon (Acipenser transmontanus). more...
