The involvement of CdhR in Porphyromonas gingivalis during Nitric Oxide stress [microarray]

(Submitter supplied) To understand the role of CdhR and its adjacent gene PG1236 in nitric oxide (NO) stress resistance, isogenic mutants P. gingivalis FLL457 (ΔPG1237::ermF), FLL458 (ΔPG1236::ermF) and FLL459 (ΔPG1236-37::ermF) were made by allelic exchange mutagenesis and their gene expression was studied under control and NO stress conditions. DNA microarray analysis of FLL457 showed that approximately 2% of the genes were up regulated and over 1% of the genes down regulated. more...

Dual RNASeq reveals NTHi-macrophage transcriptomic changes during intracellular persistence

(Submitter supplied) Nontypeable Haemophilus influenzae (NTHi) is a pathobiont which chronically colonises the airway of individuals with chronic respiratory disease. It is unclear how NTHi persists in the airway, however accumulating evidence suggests that NTHi can invade and persist within macrophages. To better understand the mechanisms of NTHi persistence within macrophages, this work developed an in vitro model of NTHi intracellular persistence using human monocyte-derived macrophages (MDM). more...

RNA Atlas of Bacterial Human Pathogens Uncovers Stress Dynamics Linked to Bacterial Infections

(Submitter supplied) Pathogenic bacteria encounter a variety of stressful host environments during infection. Their responses to meet these challenges protect them from deadly damages and aid in adaption to harmful environments. Bacterial products critical for this protection are therefore interesting as suitable targets for new antimicrobials. To shed light on the complex array of molecular pathways involved in bacterial stress responses we challenged 32 diverse human pathogenic bacteria to 11 infection related stress conditions and catalogued their transcriptomes. more...

Sustained reprioritization of metabolic pathways is associated with persistence and biofilm formation in non-typeable Haemophilus influenzae

(Submitter supplied) We used an in vitro evolved strain of nontypeable Haemophilus influenzae (NTHI) to examine the metabolic contributions to persistence RNASeq analysis identified 55 transcripts that significantly changed in amount within in vitro biofilms formed by RM33, as compared to the parental strain. Expression of genes encoding all enzymes within the tryptophan and glycogen pathways was significantly increased in biofilms formed by RM33 and the parental strains. more...

Dual RNA-sequencing of nontypeable Haemophilus influenzae and host cell transcriptomes reveals new aspects of host-pathogen interface

(Submitter supplied) Characterization of host-pathogen interactions is critical for the development of next-generation therapies and vaccines. Classical approaches involve the use of transformed cell lines and/or animal models which may not reflect the complexity and response of the human host. We reconstituted the ciliated human bronchial epithelium in vitro using primary bronchial epithelial cells to simultaneously monitor the infection-linked global changes in nontypeable Haemophilus influenzae (NTHi) and infected host epithelia gene expression by dual RNA-seq. more...

Phasevarion of non-typeable Haemophilus influenzae R2866

(Submitter supplied) H. influenzae R2866 contains a phase-variable DNA methylase modA10, by virtue of 16 tetrameric repeats present after the start codon. Gain or loss of repeats during replication by slipped-strand mispairing can result in a non-functional reading frame. Microarray analyses of wild-type and insertionally inactivated modA were performed to determine genes affected by phase variable modification.

Expression analysis of a full SlrV locus 1 knockout in Haemophilus influenzae strain PittII

(Submitter supplied) Investigation of whole genome gene expression differences between a full (four-gene) SlrV locus 1 deletion compared to the WT PittII strain. Removal of the four SlrV genes (aka macrophage survival factors : Δmsf) results in a competetive defect with the WT strain in in vitro biofilms and in the otitis media chinchilla model. It also results in an inability to persist within macrophages long-term in vitro and to escape from the middle-ear space and infect other sites in the chinchilla model.

A biphasic epigenetic switch controls immunoevasion, virulence and niche adapatation in non-typeable Haemophilus influenzae

(Submitter supplied) Non-typeable Haemophilus influenzae (NTHi) contains an N6-adenine DNA-methyltransferase (ModA), that is subject to phase variable expression (random ON/OFF switching). Five modA alleles, modA2, 4, 5, 9 and 10, account for over two-thirds of clinical otitis media isolates surveyed. Single Molecule Real Time (SMRT) methylome analysis identified the DNA recognition motifs for all five of these modA alleles. more...
