Organ - specific expression profiles of rat treated with targretin, 9-cis retinoic acid, and 4-hydroxyphenylretinamide

(Submitter supplied) A rexinoid, targretin (TRG) and two retinoids, 9-cis retinoic acid (9cRA) and 4-hydroxyphenylretinamide (4HPR) were examined for their effects on gene expression in rat mammary gland, liver, and lung tissues. The chemopreventive effects of these agents have largely been attributed to their ability to interact with retinoic acid receptors (RAR) and/or retinoid X-receptors (RXR). TRG interacts with the RXR receptors. more...

Targretin, 9-cis retinoic acid, and 4-hydroxyphenylretinamide effect on various organs

Analysis of normal livers, lungs, and mammary glands from Sprague Dawleys treated with targretin (TGR), 9-cis retinoic acid (9cRA), or 4-hydroxyphenylretinamide (4HPR). Results provide insight into the molecular basis of the varying chemopreventive anti-cancer effects of these agents.

Effects on gene expression in rat liver after administration of RXR agonists: UAB30, 4-methyl-UAB30, and Targretin

(Submitter supplied) Examination of three retinoid X receptor (RXR) agonists [Targretin (TRG), UAB30, and 4-methyl-UAB30 (4-Me-UAB30)] showed that all inhibited mammary cancer in rodents and two (TRG and 4-Me-UAB30) strikingly increased serum triglyceride levels. Agents were administered in diets to female Sprague-Dawley rats. Liver RNA was isolated and microarrayed on the Affymetrix GeneChip Rat Exon 1.0 ST array. Statistical tests identified genes that exhibited differential expression and fell into groups, or modules, with differential expression among agonists. more...

Connecting ligand-induced retinoid X receptor structural dynamics to rexinoid agonist potency

(Submitter supplied) Retinoid X receptors (RXR) are nuclear transcription factors that partner with other nuclear receptors to regulate numerous physiological processes. Although RXR represents a valid therapeutic target, only a few RXR-specific ligands (rexinoids) have been identified, in part due to the lack of clarity on how rexinoids selectively modulate RXR response. Previously, we showed that rexinoid UAB30 potentiates all-trans-retinoic acid (ATRA) signaling in human keratinocytes by stimulating ATRA biosynthesis. more...
