Gene expression analysis of immune privileged site-associated DLBCL: The effect of HLA class II downregulation

(Submitter supplied) Loss of human leukocyte antigen (HLA) expression on tumor cells is frequent in diffuse large B cell lymphoma (DLBCL) arising in immune-privileged sites such as the testis and central nervous system and is associated with small homozygous deletions of HLA-DQ/DR and larger hemizygous deletions of the major histocompatibility complex (MHC) region. To better understand the significance of downregulation of HLA class II expression in relation to the homozygous and hemizygous deletions, we analyzed global gene expression patterns in a series of 26 testicular diffuse large B-cell lymphomas (DLBCL) after characterization of these deletions. more...

Testicular diffuse large B cell lymphoma

Analysis of testicular diffuse large B cell lymphoma (DLBCL) tissues with small homozygous HLA-DQ/HLA-DR deletions, larger hemizygous MHC complex deletions, or without deletions. Results provide insight into the significance of the downregulation of HLA class II expression in testicular DLBCL.

Genomic alterations and gene expression in primary diffuse large B cell lymphomas of immune privileged sites

(Submitter supplied) Primary diffuse large B cell lymphomas of different immune-privileged sites (IP-DLBCL) share many clinical and biological features, such as a relatively poor prognosis, preferential dissemination to other immune-privileged sites and deletion of the HLA region, which suggests that IP-DLBCL represents a separate entity. To further investigate the nature of IP-DLBCL, we investigated site-specific genomic aberrations in 16 testicular, 9 central nervous system (CNS) and 15 nodal DLBCL using array-CGH. more...

Gene expression analysis of testicular DLBCL: ABC or GCB subtype?

(Submitter supplied) Diffuse large B cell lymphomas (DLBCL) constitute a heterogeneous group of lymphomas in which germinal center B cell-like and activated B cell-like subtypes can be discerned based on pathology, clinical presentation and gene expression patterns. Testicular DLBCL form an immune-privileged site-related subgroup of DLBCL with an unfavorable prognosis. We used cDNA microarray analysis, immunohistochemistry for CD10, Bcl6 and MUM1, and somatic hypermutation analysis of the immunoglobulin heavy chain gene rearrangements to determine the subtype of primary testicular DLBCL. more...

Testicular diffuse large B cell lymphoma: various subtypes

Analysis of primary testicular diffuse large B cell lymphomas (DLBCLs) and nodal DLBCLs. DLBCLs are classified as germinal center B cell-like (GCB), activated B cell-like, or undetermined. GCB is associated with a favorable prognosis. Results provide insight into the nature of testicular DLBCL.

Comparative whole genome profiling of post-transplant lymphoproliferative disorders and diffuse large B-cell lymphomas.

(Submitter supplied) Post-transplant lymphoproliferative disorders (PTLD) are a major complication of solid organ transplantation and represent a cause of severe morbidity and mortality among transplanted patients. Apart from EBV infection, knowledge of the pathogenesis of monoclonal PTLD is limited. Powerful analysis techniques, such as whole genomic DNA profiling (arrayCGH), can improve our understanding of PTLD pathogenesis. more...

CGH profiling of 87 indolent non-hodgkin’s lymphoma (NHL)

(Submitter supplied) BACKGROUND AND OBJECTIVES: Low-grade B-cell lymphomas include several subtypes of tumors with different degrees of histological, biological or clinical features. Differential diagnosis is frequently compromised by the lack of specific cytogenetic or molecular features. As a consequence, therapies remain in many lymphoma types largely based in common protocols with largely variable success. Our objectives were to describe and to compare the genomic profile of a series of samples from the most prevalent low-grade lymphoma subtypes; all of them systematically analyzed with the same approach. more...

High expression of SerpinA1 and SerpinA3 in HLA-positive cervical cell carcinomas

(Submitter supplied) In cervical cancer, an important mechanism by which tumour cells escape immune surveillance is loss of HLA class I, enabling tumours to evade recognition and lysis by cytotoxic T lymphocytes. Some tumours, however, escape from immune surveillance without accumulating defects in antigen presentation. We hypothesized that tumours with no or partial loss of HLA class I develop alternative mechanisms to prevent immune surveillance. more...
