RNAi analysis of the FBP transactivator family

(Submitter supplied) The FBP (FUSE Binding Protein) transcription factor family is composed of three highly related members; FBP/FBP1, FBP2, and FBP3. While they all share distinct modular protein structures and nucleic acid-binding specificities, their transactivator strength is graded and they also differ in their interaction with their protein partners. Especially from the extensive nuclear co-localization of these molecules, we anticipated that they co-regulate a set of common target genes. more...

Far-upstream element binding protein inactivation

FIR inhibition in HCC cells (Hep3B)

(Submitter supplied) siRNA-mediated inhibition compared to untreated cells and cells transfected with nonsense siRNA Together with far upstream element binding protein (FBP), the FBP-interacting repressor (FIR) represents a molecular tool for the transcriptional fine tune regulation of target genes. Previous evidence indicates that strong overexpression of FBP in human hepatocellular carcinoma (HCC) supports tumor growth and correlates with poor patient prognosis. more...

Global effect of JTV or JTV-Alt overexpression in HeLa cells

(Submitter supplied) JTV is not only a structural component of multi-aminoacyl-tRNA synthetase complex, but also a regulator of FBP that in turn controls c-Myc expression. In order to identify JTV target genes other than FBP and c-Myc, JTV or JTV-Alt (alternatively spliced form of JTV, enriched in the nucleus) protein was over-expressed in HeLa cells and global gene expression changes were monitored using cDNA expression microarray.
