CXCL4 induces a unique transcriptome in monocyte-derived macrophages

(Submitter supplied) Human blood monocytes were differentiated over six days with either 100 ng/ml M-CSF or 1 umol/l CXCL4 In atherosclerotic arteries, blood monocytes differentiate to macrophages in the presence of growth factors like macrophage colony-stimulation factor (MCSF) and chemokines like platelet factor 4 (CXCL4). To compare the gene expression signature of CXCL4-induced macrophages with MCSF-induced macrophages or macrophages polarized with IFN-γ/LPS (M1) or IL-4 (M2), we cultured primary human peripheral blood monocytes for six days. more...

Expression Data From HCMV-Infected Human Monocytes Study 2

(Submitter supplied) Human cytomegalovirus induces a pro-inflammatory monocyte following infection. To begin to address how HCMV induces these rapid changes in infected monocytes, we examined the transcriptome of infected monocytes. Global transcriptional profiling using cDNA microarrays revealed a significant number of pro-inflammatory genes were upregulated within 4 hours post infection. Keywords: Disease State

Mesenchymal stromal cells perturb human monocyte in vitro differentiation into dendritic cells

(Submitter supplied) Monocytes cultured in media containing GM-CSF and IL4 for 5 days normally differentiate into immature dendritic cells which, upon further stimulation with LPS for an additional 2 days, acquire a mature phenotype. In the present study, we evaluated the effect of human cord-blood mesenchymal stem cells (MSC) on human monocyte differentiation into immature and mature DCs by examining the trancriptional profile of the cells that were obtained after both the differentiation and maturation stages.

Homogeneous monocytes and macrophages from hES cells following coculture-free differentiation in M-CSF and IL-3.

(Submitter supplied) In the present study we aimed at producing homogeneous populations of monocytes and macrophages from human embryonic stem (hES) cells. Human embryonic stem cell lines KCL001, KCL002, and HUES-2 were differentiated into monocytes by coculture-free differentiation with two growth factors using a three-step method. The method involved embryoid body (EB) formation in hES media, directed differentiation with macrophage colony-stimulating factor and interleukin (IL)-3, and harvest of nonadherent monocytes from the culture supernatants. more...

Colony stimulating factor-1 receptor signaling networks inhibit macrophage inflammatory responses by induction of microRNA-21

(Submitter supplied) Macrophage polarization between the M2 (repair, pro-tumorigenic) and M1 (inflammatory) phenotypes is seen as a continuum of states. The detailed transcriptional events and signals downstream of CSF-1R that contribute to amplification of the M2 phenotype and suppression of the M1 phenotype are largely unknown. Macrophage CSF-1R pTyr-721 signaling promotes cell motility and enhancement of tumor cell invasion in vitro. more...

Extensive overlap of the STAT6 and RXR cistromes in the active enhancer repertoire of human CD14+ monocyte-derived differentiating macrophages

(Submitter supplied) Macrophages are able to differentiate into classically polarized (M1) or alternatively polarized (M2) states upon encountering pro-inflammatory cytokines such as interferon (IFN) g or anti-inflammatory cytokines such as interleukin (IL) -4/IL-13, respectively. Moreover, macrophages are known to regulate lipid metabolism via multiple members of the nuclear hormone receptor family, including the retinoid X receptors (RXR). more...

Gene expression profile during monocytes to macrophage differentiation

(Submitter supplied) Macrophages play a critical role in the pathogenesis of many diseases, including rheumatoid arthritis, inflammatory bowel disease and atherosclerosis. Monocytes recruited into tissues from peripheral blood differentiate into macrophages. There is limited data concerning the global changes in the expression of genes during monocyte to macrophage, and how the patterns of change identify the mechanism contributing to differentiation or macrophage function. more...

RNA-seq of human peripheral blood mononuclear cells

(Submitter supplied) Inflammatory cytokine responses to activation of innate immunity differ between individuals, yet the genomic and transcriptomic determinants of inflammatory responsiveness are not well understood. We hypothesized that mRNA and lincRNA expression is modulated in disease-relevant tissues in humans in vivo during inflammation, and differs between individuals with divergent evoked inflammatory responses. more...

Similarities and differences between human activated macrophages II

(Submitter supplied) We compare, in human macrophages, stimuli considered to induce M1 or M2 type of activation, also termed classical and alternative activation. This dataset highlights key differences between the M1 and M2 stimuli pointing to the need of a revision of the paradigm. Macrophages were left untreated or stimulated with IL-4 and IL-13 (prototypic M1 stimuli), Dexamethasone (Glucocorticoid receptor agonist) which has been hitherto considered M2, IL-10 which also falls under the M2 umbrella, and LPS and Interferon Gamma considered M1 stimuli. more...

Genome-wide analysis of mouse macrophages stimulated with IL-4 (Biogel and thioglycollate macrophages) (Affymetrix)

(Submitter supplied) Analysis of alternative activation of macrophages at gene expression level. The study forms part of a wider study where we compare the effects of IL-4 in different human and mouse macrophages. Our results support the notion that in vitro culture conditions greatly affect the macrophage response to IL-4. Total RNA obtained from Thioglycollate and Biogel elicited peritoneal macrophages exposed to 20 ng/ml of IL-4 for 18 hours.
