Gene expression profiling of trophoblast cells

(Submitter supplied) Invasion of cytotrophoblasts into uterine tissues is essential for placental development. To identify molecules regulating trophoblast invasion, mRNA signatures of purified villous (CTB, poor invasiveness) and extravillous (EVT, high invasiveness) trophoblasts isolated from first trimester human placentae and villous explant cultures, respectively, were compared using GeneChip analyses yielding 991 invasion/migration related transcripts. more...

Gene expression profiles of trophoblast cell lines

(Submitter supplied) Invasive extravillous trophoblasts (EVTs) of the human placenta are critically involved in successful pregnancy outcome since they remodel the uterine spiral arteries to increase blood flow and oxygen delivery to the placenta and the developing fetus. To gain more insights into their biological role different primary cell culture models are commonly utilised. However, access to early placental tissue may be limited and primary trophoblasts rapidly cease proliferation in vitro impairing genetic manipulation. more...

LOXs in invasive trophoblasts

(Submitter supplied) Human placental development is characterized by invasion of extravillous cytotrophoblasts (EVCTs) into the uterine wall during the first trimester of pregnancy. Peroxisome proliferator-activated receptor gamma (PPARG) plays a major role in placental development, and activation of PPARG by its agonists results in inhibition of EVCT invasion in vitro. To identify PPARG target genes, microarray analysis was performed using GeneChip technology on EVCT primary cultures obtained from first-trimester human placentas. more...

Gene expression signatures of HO-1 in BeWo cells

(Submitter supplied) Heme Oxygenase-1 (HO-1) is expressed in many cancers and influences the growth, survivall and metastasis of tumors, however, the molecular mechanisms remains largely unknown. To identify a common mechanism of action of HO-1 in cancer, we studied the global effect of HO-1 on the transcriptome of multiple tumors. Genome-wide expression profiling of HO-1 expressing versus HO-1 silenced cancer cells and a further data mining analysis of the preexisting expression database of 190 human tumors of 14 cancer types led us to identify 14 genes, the expression of which correlated firmly and universally with that of HO-1 (P < 0.001). more...

Hypoxia induced HIF-1/HIF-2 activity alters trophoblast transcriptional regulation and promotes invasion

(Submitter supplied) Reduced or absent cytotrophoblast invasion of the maternal uterine spiral arteries is a common clinical finding in studies of pregnancies complicated by preeclampsia, suggesting that the mechanisms behind invasion of these cells is perturbed. The placenta initially develops in a low oxygen environment of 1-2% oxygen until after the 10th week of pregnancy. During this time oxygen concentration exerts a major influence over trophoblast activity and, in vitro, hypoxia inducible factors are proposed to be one of many key regulators of first trimester trophoblast behaviour. more...

RNA-Seq analysis in human trophoblast stem cells (CT27) upon depletion of Hippo signaling Component TAZ (WWTR1)

(Submitter supplied) The hippo signaling cofactor TAZ (Also known as WWTR1) regulates expression of genes upon interaction with TEAD transcription factors. Among the TEAD transcription factors, TEAD4 is implicated in regulation of the self-renewal process of human trophoblast stem cells. However, we have a poor understanding about the contribution of TAZ, a co-factor of TEAD4, in that process. Thus, the goal of this study is to define the importance of TAZ in orchestrating global gene expression program in human trophoblast stem cells. more...

Preeclampsia is associated with reduced ISG15 levels impairing extravillous trophoblast invasion.

(Submitter supplied) Preeclampsia (PE), a major cause of excess maternal and perinatal morbidity and mortality, has a worldwide prevalence of 2-8%. Excess inflammation with immune maladaptation in the maternal decidua causes shallow trophoblast invasion, resulting in incomplete spiral artery transformation that are characteristic of PE. To mimic the separate and combined paracrine effects of IL-11 and IL-6 on trophoblasts in normal versus preeclamptic pregnancies, primary cytotrophoblast (CTB) cultures treated with IL-11 or IL-6 or IL-11+IL-6 were analyzed by whole genome microarray. more...

Wide ranging DNA methylation differences of primary trophoblast cell populations and derived-cell lines

(Submitter supplied) Difficulties associated with long term culture of primary trophoblasts have proven to be a major hurdle in their functional characterization. In order to circumvent this issue, several model cell lines have been established over many years using a variety of different approaches. Due to their differing origins, gene expression profiles, and behavior in vitro, different model lines have been utilized to investigate specific aspects of trophoblast biology. more...

Single cell trajectory modeling identifies a primitive trophoblast state defined by BCAM enrichment

(Submitter supplied) The establishment and function of the human placenta is dependent on specialized cells called trophoblasts. Unfortunately, little is known about the cellular and molecular processes controlling human trophoblast stem cell maintenance and differentiation into mature trophoblast sub-populations/cell states. To address this, we here report transcriptomic data from n=7 first trimester human placental tissues, n=3 regenerative human trophoblast stem cell (hTSC) derived trophoblast organoids, and n=3 EVT-differentiated hTSC derived organoid cultures at single-cell resolution. more...
