Inherent gene signature of stage II and III colorectal cancer leads in individual recurrence

(Submitter supplied) Background & Aims. The current staging system for colorectal cancer (CRC) based on TNM classification allows prediction of potential recurrence. However, it does not necessarily make reliable personalized prediction of prognosis. In this paper we describe combination of clinicopathological data and gene signature of dissected tumor specimen with stage II and III CRC patients would improve the situation.. more...

Gene level expression profiling of colorectal cancer tissue samples

(Submitter supplied) By the use of whole genome transcription analysis, we aimed to develop a gene expression classifier to increase the likelihood of identifying stage II colorectal cancer (CRC) samples with a poor prognostic outcome. Gene expression measurement were measured by the GeneChip® Human Exon 1.0 ST Arrays from Affymetrix.

Exon level expression profiling of colorectal cancer tissue samples (validation sample series).

(Submitter supplied) Colorectal cancer is a heterogeneous disease molecularly characterized by inherent genomic instabilities, chromosome instability and microsatellite instability. In the present study we propose transcriptome instability as an analogue to genomic instability on the transcriptome level. Exon microarray data from two independent series of altoghether 160 colorectal cancer tissue samples was used for global alternative splicing detection using the FIRMA algorithm (aroma.affymetrix). more...

Clinical Significance of Osteoprotegerin Expression in Human Colorectal Cancer

(Submitter supplied) Purpose: This study aimed to identify a novel biomarker or a target of treatment for colorectal cancer (CRC). Experimental Design: The expression profiles of cancer cells in 104 patients with CRC were examined using laser microdissection and oligonucleotide microarray analysis. Overexpression in CRC cells especially in patients with distant metastasis was a prerequisite to select candidate genes. The mRNA expression of candidate gene was investigated by quantitative reversetranscription polymerase chain reaction (RT-PCR) in 77 patients as a validation study. more...

Gene expression signature and the prediction of recurrence in stage III colorectal cancer by DNA microarray

(Submitter supplied) Samples were taken from colorectal cancers in surgically resected specimens in 36 colorectal cancer patients. The expression profiles were determined using Affymetrix Human Genome U133 Plus 2.0 arrays. Comparison between the sample groups allow to identify a set of discriminating genes that can be used for molecular markers for predicting recurrence. Keywords: repeat

Expression Changes Relevant for Apoptosis in K562 Cells Co-Treated with Amifostine and Imatinib

(Submitter supplied) We had previously demonstrated an increased proapoptotic effect of Imatinib (STI571) in combination with Amifostine (AMI) in K562 cell line. In this study we used genomic scale gene expression profiling to monitor changes at transcriptional level in K562 cells during the treatment with AMI+STI571. We identified a transcriptional repressor of survival genes, known as BTF, which triggers a pro-apoptotic signal, potentially helpful to overcome the resistance to STI571. more...

Comparative study of gene expression in human colorectal cancer tissues and normal mucosa

(Submitter supplied) The purpose of our study was to better understand the genetic mechanism of oncogenesis for human colorectal cancer and to identify potential new tumour markers of use in clinical practice. Results: Comparing cancerous tissues with normal colonic mucosa we identified 584 known genes (3%) differentially expressed to a significant degree (p<0.001). Many of the transcripts that were more abundant in tumours than in non-neoplastic tissues appear to reflect important events for colon carcinogenesis. more...

Classification of serrated colorectal tumors

(Submitter supplied) Serrated adenocarcinomas are morphologically different from conventional adenocarcinomas. The serrated pathway has recently been proposed to represent a novel mechanism of colorectal cancer (CRC) formation. However, whether they are biologically different and truly form a distinct subclass of CRC, is not known. This study shows that the gene expression profile of serrated and conventional CRCs differs from each others and that serrated CRCs are not only morphologically novel, but also biologically distinct subclass of CRC. more...

Expression data from healthy controls and early stage CRC patient's tumor

(Submitter supplied) Metastasis is the major cause of cancer mortality. Up to 25% of early stage sporadic colorectal cancer (CRC) patients succumb to metastasis after curative surgery. We used microarrays to detail gene expression and identified a metastasis-prone signature for early stage CRC. Keywords: RNA expression
