Differential expression of rituximab responders vs. non responders on 3 different blood cell types

(Submitter supplied) New and effective therapeutical options are available for the treatment of Rheumatoid Arthritis. One of such treatments is rituximab, and chimeric anti-CD20 antibody that selectively depletes the CD20+ B cell subpopulation. Similar to established anti-TNF alpha therapies, there is a subgroup of RA patients that do not experience significant clinical response. Therefore, one of the major necessities in actual RA therapeutical management is to identify reliable predictors of the response to this therapies. more...

mRNA expression profile in peripheral blood cells of RA patients following treatment with an anti-TNFa mAb, infliximab

(Submitter supplied) Objectives: We monitored the mRNA expression profiles of peripheral blood cells during treatment with a tumor necrosis factor-a inhibitor, infliximab, in patients with rheumatoid arthritis (RA). Using a DNA microarray analysis, we demonstrated a unique set of genes, with distinct baseline and post-treatment changes in expression between responders and non-responders to infliximab treatment. Methods: Using a customized low-density cDNA microarray and a reliable data collection system, we monitored the mRNA expression profiles of whole blood cells from 18 RA patients before and after the infusion of infliximab for up to 22 weeks. more...

Efficacy of anti-TNF biologic agent, infliximab, for RA patients using transcriptome analysis of white blood cells

(Submitter supplied) Introduction of biologics, such as infliximab, to the therapy of rheumatoid arthritis (RA) patients has revolutionized the treatment of this disease. However, biomarkers for predicting the efficacy of the drug at an early phase of treatment for selecting real responders have not been found. We here present predictive markers based on a thorough transcriptome analysis of white blood cells from RA patients. more...

Gene expression profiling of rheumatoid arthritis synovial tissues

(Submitter supplied) Spots were selected that had an expression level higher than 1.6 the local background. The use of a common reference sample allows the comparison of the relative expression levels across the tissue samples. All genes were expressed relative to their median expression level across the arrays of the same microarray platform, allowing us to combine the data from the 2 platforms. Data from Lymphochip and Stanford Human cDNA arrays was combined per patient. more...

Identification of markers of responsiveness in rheumatoid arthritis patients

(Submitter supplied) Despite the increasing of available drugs in RA, the overall rate of non response is 30 to 40% of treated patients. To optimize the drug prescription and better elaborate the therapeutic strategies, gene profiling in PBMCs was used to predict the responsiveness to anakinra. Thirty two patients treated by anakinra (100mg/day s.c.) and méthotrexate, were categorized as responders whenever a change of Disease Activity Score 28 > 1.2 was obtained at 3 months. more...

Effects of Rituximab on global gene expression profiles in the RA synovium

(Submitter supplied) Objective: Rituximab displays therapeutic benefits in the treatment of rheumatoid arthritis (RA) patients resistant to TNF blockade. However, the precise role of B cells in the pathogenesis of RA is still unknown. In this study we investigated the global molecular effects of rituximab in synovial biopsies obtained from anti-TNF resistant RA patients before and after administration of the drug. Methods: Paired synovial biopsies were obtained from the affected knee of anti-TNF resistant RA patients before (T0) and 12 weeks after initiation of rituximab therapy (T12). more...

Rheumatoid arthritic synovium response to rituximab

Analysis of synovial biopsy samples from anti-TNF resistant rheumatoid arthritis (RA) patients 12 wks after the start of rituximab (RTX) therapy. RTX is a mAb against CD20, expressed on subsets of B cells. Results provide insight into how RTX-induced B cell depletion interferes with RA pathogenesis.

Genome-wide transcription analysis of whole blood prior to rituximab treatment in relation to clinical response

(Submitter supplied) B cell depletion therapy is efficacious in RA patients failing on TNF blocking agents. However, approximately 40-50% of the rituximab-treated RA patients have a poor response. We investigated wheter baseline gene expression levels can discriminate between clinical nonresponders and responders to rituximab

Gene expression profiles of human promyelocytic leukemia cell lines exposed to six volatile organic compounds

(Submitter supplied) The gene expression profiles of HL-60 cells were determined for two doses (IC20 and IC50) of three chemicals (ethylbenzene, tricholoretylene and dichloromethane) and for the IC50 doses of 3 other chemicals (benzene, toluene and o-xylene) at a 3 h time point. Assays were performed in triplicate for each chemical/dose, and individual gene-expression profiles were determined for each chemical at the tested concentration doses.

The relationship between the gene expression profile in the synovium of RA patients at baseline and the clinical response to infliximab treatment

(Submitter supplied) Background: Although the use of TNF inhibitors has fundamentally changed the way rheumatoid arthritis (RA) is treated, not all patients respond well. It is desirable to facilitate the identification of responding and non-responding patients prior to treatment, not only to avoid unnecessary treatment but also for financial reasons. In this work we have investigated the transcriptional profile of synovial tissue sampled from RA patients before anti-TNF treatment with the aim to identify biomarkers predictive of response. more...

Discovery of new candidate genes for rheumatoid arthritis through integration of genetic association data with expression pathway analysis

(Submitter supplied) Purpose: The goal of the study was to integrate verified signals from previous genetic association studies with gene expression and pathway analysis for discovery of new candidate genes and signalling networks, relevant for rheumatoid arthritis (RA). Method:RNA-seq based expression analysis of 377 genes from previously verified RA-associated loci was performed in blood cells from 5 newly diagnosed, non-treated RA patients, 7 patients with treated RA and 12 healthy controls. more...

Expression data from active rheumatoid arthritis patients and healthy control.

(Submitter supplied) Rheumatoid arthritis (RA) is a systemic autoimmune disease and its underlying molecular mechanisms are still poorly understood. Previously a CD4 T-cell microarray study has only focused arthritis patients. We aimed to compare the molecular profiles of active RA versus healthy control in CD4 T cells. We used microarrays to detail the global programme of gene expression in active RA and healthy control.
