Gene expression profiling of human mammary artery media during aging

(Submitter supplied) Our goal was to better understand the genes and pathways implicated during arterial media aging. We therefore compared the gene expression profile of human mammary artery media between Young (<60, 8 patients) and old (>75, 7 patients) patients. The microarray data, generated using the Illumina sentrix Ref8-V2 platform, was normalized and analyzed using the IlluminaGUI package (Eggle and Schultze, 2007).

Whole Human Genome Oligo Microarray

(Submitter supplied) Human expression microarray analysis of the 11 samples that you submitted. Total RNA from each sample was quantified using the NanoDrop ND-1000 and the RNA integrity was assessed using standard denaturing agarose gel electrophoresis. For microarray analysis, Agilent Array platform was employed. The sample preparation and microarray hybridization were performed according to the manufacturer’s standard protocols. more...

Altered modulation of WNT-β-catenin and PI3K/Akt pathways in IgA nephropathy

(Submitter supplied) To uncover new molecular mechanisms involved in IgAN pathogenesis, we compared the genomic profiles of 12 IgAN patients with 8 healthy subjects, We included addiotional disease controls 3 FSGS(Focal Segmenal glomerulosclerosis) patients and 3 membrano proliferative glomerulonephritis type I (MPGN-I) controls in our study to evaluate if the WNT-β-catenin and PI3K/Akt pathways were specfic to IgAN.

Redifferentiation of adult human β cells expanded in vitro by inhibition of the WNT pathway

(Submitter supplied) In vitro expansion of adult human islet β cells is an attractive solution for the shortage of tissue for cell replacement therapy of type 1 diabetes. Using a lineage tracing approach, we have demonstrated that β-cell-derived (BCD) cells rapidly dedifferentiate in culture and can proliferate for up to 16 population doublings. Dedifferentiation is associated with changes resembling epithelial-mesenchymal transition (EMT). more...

Suppressed cellular senescence mediated by T-box3 in aged gastric epithelial cells may contribute to aging-related carcinogenesis

(Submitter supplied) Aging is a risk factor of carcinogenesis in various organs, and abnormal aging of the stem cells is considered responsible for carcinogenesis. Recent advances in organoid culturing system have made it viable to investigate mini organs developed from tissue stem cells. In this study, we aimed to investigate the implications of aging stem cells for gastric carcinogenesis. We found that gastric organoids of aged mice grew larger in size and proliferated vigorously compared to that of young mice. more...
