Expression data from wild type or FIZZ2 knockout murine lungs

(Submitter supplied) To study the possible fibrotic role of FIZZ2, bleomycin was used to induce pulmonary fibrosis in wild type and FIZZZ2 knockout mice, lungs were then harvested and processed for RNA isolation. We used microarrays to detail the global gene expression regulated by FIZZ2 during fibrotic process.

Expression data from mouse lung epithelium after allergen challenge

(Submitter supplied) Changes occur in the lung epithelium after allergen challenge that may give insight into asthma pathogenesis and we sought to identify novelepithelial genes induced by allergen exposure. We used microarrays to identify up-regulated genes induced by allergen challenge in lung epithelium.

Lung epithelium response to fungal allergen Alternaria

Analysis of bronchial brushings of lung epithelium from C57BL/6 females after Alternaria challenge. Alternaria allergen is associated with severe asthma, including life-threatening exacerbations. Results provide insight into the molecular basis of the acute innate airway response to Alternaria.

Gene expression analysis of lung fibroblasts in bleomycin-induced pulmonary fibrosis

(Submitter supplied) We conducted fibroblast-specific transcriptome analysis by next generation sequencing in order to investigate qualitative change and activation signatures of lung fibroblasts in bleomycin-induced pulmonary fibrosis. Lung fibroblasts were identified by using reporter mice of collagen-α2(I), in which collagen I-producing fibroblasts were labeled with EGFP. Lungs were dissociated with protease sollution, and single cell suspension were stained with lineage markers (Ter119, CD45, CD31, EpCAM). more...

MMP1 and MMP7 as potential peripheral blood biomarkers in Idiopathic Pulmonary Fibrosis

(Submitter supplied) Idiopathic pulmonary fibrosis (IPF) is a chronic progressive fibrotic lung disease associated with significant morbidity and mortality. We identified a combinatorial signature of 5 proteins that was sufficient to distinguish IPF patients from controls; of these proteins MMP7 and MMP1 exhibited significantly higher values in both peripheral blood concentrations and lung tissue gene expression, suggesting their role as true biomarkers. more...

Development of gene expression signatures for Mmp19 regulation of fibroblast phenotype in mouse lungs

(Submitter supplied) We have employed whole genome microarray expression profiling as a discovery platform to identify genes with the potential to distinguish Mmp19 regulation of fibroblast phenotype changes in mouse lungs. Pulmonary fibrosis was induced by bleomycin at 0.08 u in 50ul of saline. At 21st day the mice were sacrificed and mouse lung fibroblasts were isolated and cultured in FBM plus additives following Lonza's portocol. more...

Epiregulin is a dendritic cell-derived EGFR ligand that maintains skin and lung fibrosis

(Submitter supplied) Immune cells are fundamental regulators of extracellular matrix (ECM) production by fibroblasts and have important roles in determining extent of fibrosis in response to inflammation. Although much is known about fibroblast signaling in fibrosis, the molecular signals between immune cells and fibroblasts that drive its persistence are poorly understood. We therefore analyzed skin and lung samples of patients with diffuse cutaneous systemic sclerosis, an autoimmune disease that causes debilitating fibrosis of the skin and internal organs. more...

Resistin-like molecule α provides vitamin A-dependent antimicrobial protection of the skin

(Submitter supplied) The skin is the human body’s largest organ and is in contact with a diverse community of microorganisms that includes both resident and pathogenic bacteria. Skin immune defenses include the production of antimicrobial proteins that kill bacteria directly. However, we still have an incomplete understanding of how skin antimicrobial proteins promote homeostasis with resident bacterial communities and limit infection. more...
