Neurotoxicogenomic investigations to assess mechanisms of action of the munitions constituents RDX and 2,6-DNT in Northern bobwhite (Colinus virginianus) (part 5 of 10)

(Submitter supplied) Munitions constituents (MCs) including hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX), 2,4,6-trinitrotoluene (TNT), and TNT derivatives are recognized to elicit aberrant neuromuscular responses in many species. The onset of seizures resulting in death was observed in the avian model Northern bobwhite after oral dosing with RDX beginning at 8 mg/kg/day in subacute (14 days) exposures, whereas affective doses of the TNT derivative, 2,6-dinitrotoluene (2,6-DNT), caused gastrointestinal impacts, lethargy, and emaciation in subacute and subchronic (60 days) exposures. more...

Rat exposure to RDX (3mg/kg or 18mg/kg; 0, 4, 24, 48 hr)

(Submitter supplied) RDX (Hexahydro-1,3,5-trinitro-1,3,5-triazine) is a synthetic, high-impact, relatively stable explosive that has been in use since WWII. Exposure to RDX can occur either occupationally or through ordnance that lays unexploded on training ranges. The toxicology of RDX is dominated by acute tonic-clonic seizures at high doses, which remit when exposure is removed and internal RDX levels decrease. Sub-chronic studies have revealed few other toxic effects. more...

Systems Toxicology Identifies Mechanistic Impacts of 2-amino-4,6-dinitrotoluene (2-ADNT) Exposure in Northern Bobwhite

(Submitter supplied) A systems toxicology investigation comparing and integrating transcriptomic and proteomic results was conducted to pursue a holistic understanding of the effects of the explosives degradation product 2-amino-4,6-dinitrotoluene (2A-DNT) dosing on the health of the wildlife bird model, Northern bobwhite (Colinus virginianus). A subchronic 60d toxicology bioassay was leveraged where both sexes were dosed via daily gavage with 0, 3, 14, or 30 mg/kg-d 2-ADNT. more...

Profiling differentially expressed earthworm genes in response to TNT, RDX, or TNT+RDX exposure.

(Submitter supplied) To understand molecular mechanisms of the joint effects of 2,4,6-trinitrotoluene (TNT) and hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX), both widely used ordnance compounds, we constructed a microarray consisting of 4,032 cDNA isolated from the earthworm Eisenia fetida using the suppressive subtractive hybridization technique. Worms were exposed to TNT-, RDX-, or TNT+RDX-spiked soil for 28 days (TNT 50 mg/kg, RDX 30 mg/kg). more...

Transcriptomics Provides Mechanistic Indicators of Mixture Toxicology for IMX-101 and IMX-104 Formulations in Fathead Minnows (Pimephales promelas) [IMX-104]

(Submitter supplied) Abstract — Within the US military, new insensitive munitions (IMs) are rapidly replacing conventional munitions improving safety from unintended detonation. Toxicity data for IM chemicals are expanding rapidly, however IM constituents are typically deployed in mixture formulations, and very little is known about their mixture toxicology. In the present study we sought to characterize the mixture effects and toxicology of the two predominant IM formulations IMX-101 and IMX-104 in acute (48 h) larval fathead minnow (Pimephales promelas) exposures. more...
