Silencing of Irf7 pathways in breast cancer cells promotes bone metastasis through immune escape mechanisms

(Submitter supplied) Breast cancer metastasis to bone is a critical determinant of long-term survival after treatment of primary tumors. We used a mouse model of spontaneous bone metastasis to determine new molecular mechanisms. Differential transcriptome comparisons of primary and metastatic tumor cells revealed that a substantial set of genes suppressed in bone metastases were highly enriched for promoter elements for the type I interferon (IFN) regulatory factor, Irf7, itself suppressed in mouse and human metastases. more...

Type I interferon gene expression response to chemotherapy

(Submitter supplied) Neoadjuvant and adjuvant chemotherapies provide survival benefits to breast cancer patients by delaying distant relapses or inducing cure, but the mechanism is not fully clear. Here, we performed a gene expression analysis to demonstrate that estrogen-receptor (ER)-negative murine 4T1 mammary adenocarcinoma cells treated by chemotherapy in vitro activate the IFNß/IFNAR/IRF7 pathway to elicit a T cell-dependent immune response that maintains cancer cells in a dormant state in vivo.

Gene expression profile of peripheral blood lymphocytes: comparison between melanoma patients and healthy controls

(Submitter supplied) We focused on the major peripheral blood lymphocyte populations that may be involved in anti-tumor responses and negatively impacted by cancer, specifically CD8 T cells, CD4 T cells, B cells and CD56dim natural killer cells. The pure cell subsets were stringently sorted by flow cytometry from PBMC samples. Gene expression profiles of these cell populations from melanoma patients were compared to healthy controls. more...

Metastatic melanoma: peripheral blood lymphocytes

Analysis of sorted peripheral blood lymphocytes (CD8 T cells, CD4 T cells, B cells, NK cells) from patients with melanoma. These subpopulations are involved in antitumor responses and negatively impacted by cancer. Results provide insight into molecular mechanisms of immune dysfunction in cancer.

Zka virus (ZIKV) persistently infects primary human brain microvascular endothelial cells

(Submitter supplied) ZIKV alters transcriptional responses of infected cells to avoid immune detection. We have compared the transcriptional responses of ZIKV-infected hBMECs and mock-infected hBMECs. Examination of transcriptional responses of ZIKV-infected hBMECs versus mock-infected hBMECs

Transcriptomics of the traditional Japanese medicine juzentaihoto (JTX) on small intestines on germfree mice

(Submitter supplied) Juzehtaihoto, a Japanese traditional medicine has been used for the treatment of various kinds of disease or disorders in an enteric-flora dependent manner. Here, we performed transcriptome analysis using Affymetrix GeneChip on small intestine (SI) of germ-free (GF) and specific pathogen free (SPF) mice of IQI, an inbred strain established from ICR.

Comparative profiling of metastatic 4T1- versus non-metastatic Py230-based mammary tumors in an intraductal model for triple-negative breast cancer.

(Submitter supplied) Purpose: Compare the tumor outgrowth and immunology of an immunocompetent 4T1- and Py230-based intraductal model for triple-negative breast cancer (TNBC). Methods: BALB/c-derived 4T1 and C57BL/6-derived Py230 mammary tumor cells were side-by-side intraductally inoculated in lactating and syngeneic mice, 4T1 and Py230 primary tumors were subsequently resected at 1, 3 and 6 weeks (w) post-inoculation (p.i.), and RNA was isolated from the 4T1 and Py230 primary tumors using in house developed protocols. more...

Transcriptomic and Epigenetic analysis of cell line derivatives from H2087 and HCC1954 cell lines

(Submitter supplied) We profiled the transcriptomes of latency-competent cells derived from the human cancer cell lines H2087 (lung adenocarcinoma) and HCC1954 (breast adenocarcinoma) in mitogen-rich and mitogen-low media (MRM and MLM, respectively). In addition, we analyzed the epigenetic landscape of these cell lines under MLM conditions.
