Characterization of cholesterol loading effect on the gene expression of bone marrow derived macrophages of AKR and DBA/2 ApoE-deficient mice

(Submitter supplied) Strain effects on the response to cholesterol loading in bone marrow derived macrophages (BMM) are not well studied. The atherosclerosis susceptible strain, DBA/2 on the ApoE deficient background and the atherosclerosis resistant AKR strains show differences in cholesterol handling when loaded with AcLDL. To understand the molecular basis for these differences, BMM from both strains were loaded with acetylated low desnisty lipoprotein (AcLDL) and gene expression quantified. more...

Increased endoplasmic reticulum stress response is involved in clopidogrel-induced apoptosis of gastric epithelial cells

(Submitter supplied) Clopidogrel is associated with a series of gastrointestinal side effects, such as bleeding complications and recurrent gastric ulcer; however, their mechanisms are largely unclear. In this study, human gene expression microarray and gene ontology analysis were utilized to evaluate the effects of clopidogrel on gene expression in human gastric epithelial cells (GES-1) Keywords: Clopidogrel; Gastric injury; GES-1 cell line

Small artery remodeling depends on the genetic background in spontaneously hypertensive rats (mRNA)

(Submitter supplied) Spontaneously hypertensive rats (SHR) have been used frequently as a model for human essential hypertension. However, both the SHR and its normotensive control, the Wistar Kyoto rat (WKY), consist of genetically different sublines. We tested the hypothesis that discrepant data in literature regarding the pathophysiology of vascular remodeling in hypertension result from the use of different rat sublines. more...

Expression data from the aorta of DBA, B6 and 129 mice

(Submitter supplied) Strain differences influence susceptibility to atherosclerosis. Apolipoprotein E-null mice on a DBA/2J genetic background (DBA-apoE) and C57BL/6 (B6-apoe) are highly susceptible to atherosclerosis in the aortic root area compared with those on a 129S6/SvEvTac background (129-apoE). To explore strain-specific differences affecting the susceptibility to atherosclerosis, we performed microarray analysis of aortic arch and root from wild type mice of each strains.

C57BL/6 C3H/HeJ apoE-/- mice - gene expression in aorta

(Submitter supplied) Inbred mouse strains C57BL/6J (B6) and C3H/HeJ (C3H) exhibit a marked difference in their susceptibility to atherosclerosis and the arterial wall has proven to be a source of the difference in atherosclerosis susceptibility. Genome-wide gene expression analysis was conducted in aortic walls of the two strains. Total RNA was extracted from aortas of 6-week-old female B6 and C3H apoE-deficient (apoE-/-) mice fed a chow or Western diet. more...

RNA sequencing profiling of the retina in C57BL/6J and DBA/2J mice: enhancing the retinal microarray datasets from GeneNetwork

(Submitter supplied) Purpose: The goal of the present study is to provide an independent assessment of the retinal transcriptome signatures of the C57BL/6J (B6) and DBA/2J (D2) mice and to enhance existing microarray datasets for accurately defining the allelic differences in the BXD recombinant inbred strains. Methods: Retinas from both B6 and D2 mice (3 of each) were used for the RNA-seq analysis. Transcriptome features were examined for both strains. more...

Molecular mechanisms vascular smooth muscle cell (VSMC) acquisition of macrophage features in response to cholesterol loading

(Submitter supplied) In this study, murine primary aortic smooth muscle cells (SMCs) were transcriptionally profiled at baseline, after 3 d of cholesterol loading, and after 3 d of subsequent cholesterol unloading with HDL treatment, to identify vascular SMC genes that are transcripionally dysregulated in response to cholesterol loading and/or unloading.

Mechanisms of urokinase plasminogen activator (uPA)-mediated atherosclerosis

(Submitter supplied) Data from clinical studies, cell culture, and animal models implicate the urokinase (uPA)/Plasminogen (Plg) system in the development of atherosclerosis and aneurysms. However, the mechanisms through which uPA/Plg stimulate these diseases are not yet defined. We used genetically modified, atherosclerosis-prone mice, including mice with macrophage-specific uPA overexpression to clarify mechanisms of uPA/Plg-accelerated atherosclerosis and aneurysm formation. more...

Ath29 congenic mice - gene expression in aorta

(Submitter supplied) A congenic mouse line was constructed by introgressing a C3H chromosome 9 region harboring Ath29 into the C57BL/6 apoE-deficient background. RNA was extracted from aorta using a QIAGEN kit . Total RNA was pooled in an equal amount from 3 mice for each group. Standard Affymetrix procedures were performed using 8ug of total RNA. Microarrays were used to detect gene expression in the aorta of Ath29 congenic mice and C57BL/6 apoE-deficient mouse strains fed a chow or western diet.
