A BRCA1 Deficient-Like Signature is Enriched in Breast Cancer Brain Metastases

(Submitter supplied) Purpose: There is an unmet clinical need for biomarkers to identify breast cancer patients who are at increased risk of developing brain metastases. The objective is to identify gene signatures and biological pathways associated with HER2+ brain metastasis. Experimental Design: Gene expression of 19 HER2+ breast cancer brain metastases was compared with HER2+ nonmetastatic primary tumors. BRCA1-related gene signature in breast cancer: the role of ER status and molecular type

(Submitter supplied) We have analyzed, using DNA microarrays, putative differences in gene-expression level between hereditary BRCA1 mutation-linked and sporadic breast cancer. Our results show that a previously reported marked difference between BRCA1-mutation linked and sporadic breast cancer was probably due to uneven stratification of samples with different ER status and basal-like versus luminal-like subtype. We observed that apparent difference between BRCA1-linked and other types of breast cancer found in univariate analysis was diminished when data were corrected for ER status and molecular subtype in multivariate analyses. more...

Understanding BRCA1 function in INK4-RB deficient tumors

(Submitter supplied) Most BRCA1-deficient BLBCs carry a dysfunctional INK4-RB pathway. Thus, we have created genetically engineered mice with Brca1 loss and deletion of p16INK4A, or separately p18INK4C, to model the deficient INK4-RB signaling in human BLBC. By using these mutant mice and human BRCA1 deficient and proficient breast cancer tissues and cells, we tested if there exists a druggable target in BRCA1 deficient breast cancers.

Multi-omics Profiling of Younger Asian Breast Cancers Reveals Distinctive Molecular Signatures

(Submitter supplied) We observed higher proportions of HER2+ and Luminal B but lower proportions of ER+ and Luminal A subtypes along with lower estrogen receptor (ER) expression in SMC compared to TCGA. Germline pathogenic mutations affecting BRCA1 or BRCA2 were found in 11% of SMC but only 5% of TCGA. TP53 was also more frequently mutated in SMC (48%) than in TCGA (32%).

RNAseq of mouse mammary luminal cells

(Submitter supplied) The mammary epithelia are mainly composed of two distinct lineages, the basal and luminal cells. In our MMTV-Cre; Brca1flox/flox mouse model, we found the Brca1 knockout mainly occurred in the luminal cells, which will lead the mammary tumorigenesis. To investigate the Brca1 deficiency mediated mammary tumorigenesis, we sorted the luminal cells from wild type mice and MMTV-Cre; Brca1flox/flox mice for RNAseq analysis.
