TM4SF20 is reduced by TGF-β in A549 cells

(Submitter supplied) To find out genes regulated by TGF-β in A549 cells, we compared gene expression of cells treated with 1ng/ml TGF-β versus non-treated cells and find out that expression of one transmembrane protein, TM4SF20, is reduced by TGF-β.

CREB3L1 Target Genes in Response to Hepatitis C Replicon Infection

(Submitter supplied) Membrane-bound transcription factor CREB3L1 undergoes Regulated Intramembrane Proteolysis (RIP) in response to Hepatitis C infection. RIP activates CREB3L1 so that it can prevent the growth of HCV infected cells through the action of downstream genes. We over-expressed a truncated form of CREB3L1 that does not require RIP to enter the nucleus. Cells over-expressing this truncated form were isolated by Fluorescence Activated Cell Sorting (FACS). more...

Host Factors with Reduced Expression in Two HCV Permissive Cell Lines as Compared to the Non-Permissive Parent Cell Line Huh7

(Submitter supplied) Drugs directly targeting Hepatitis C (HCV) are often rendered useless by the high mutation rate of the virus. Thus, we deduce that targeting of host factor that affect HCV replication may provide enhanced therapy fort HCV infection. Hepatocyte cell line Huh7 is known to be non-permissive for Hepatits C (HCV) replication. Through a method developed by the Rice laboratory (Blight, K.J., et al., J Virol, 2002), selection of a small subset of permissive hepatocytes is possible. more...

Hepatitis C virus permissive cell lines Huh7.5 and HRP1 and non-permissive parent cell line Huh7

Analysis of hepatocyte cell line Huh7, non-permissive to hepatitis C virus (HCV) replication, and permissive cell lines Huh7.5 and HRP1. Results provide insight into host factors lost in cells permissive for HCV replication.

Knockdown of transcription factor Creb3l1 in anterior pituitary mouse corticotroph cell line AtT20

(Submitter supplied) Transcription factor Creb3l1 is a non-classical ER stress molecule that is emerging as an important component for cellular homeostasis, particularly within cell-types with high peptide secretory capabilities. We have previously shown that Creb3l1 serves an important role in body fluid homeostasis through its transcriptional control of the gene coding for antidiuretic hormone arginine vasopressin in the neuropeptide rich magnocellular neurons of the supraoptic nucleus. more...

TGF-beta regulated gene transcription and protein expression in human GFAP-negative lamina cribrosa cells

(Submitter supplied) Primary open angle glaucoma (POAG) is a progressive optic neuropathy, which is a major cause of worldwide visual impairment and blindness. Pathological hallmarks of the glaucomatous optic nerve head include retinal ganglion cell axon loss and extracellular matrix (ECM) remodeling of the lamina cribrosa layer. TGF-beta is an important pro-fibrotic modulator of ECM metabolism, whose levels are elevated in human POAG lamina cribrosa tissue compared with non-glaucomatous controls. more...

GFAP-negative lamina cribrosa cell response to TGF-beta1

Analysis of glial fibrillary acidic acid (GFAP)-negative lamina cribrosa (LC) glial cell response to 10 ng/ml TGF-beta1 for 24 hours. This is a model for primary open-angle glaucoma (POAG) in which TGF-beta levels are elevated in human LC tissue.

Microarray of RNA from calvaria in WT and OASIS-/- (KO) mice

(Submitter supplied) Investigation of whole genome gene expression level changes in OASIS KO calvaria compared to wild-type calvaria. To gain further insight into the potential mechanisms underlying the defective bone formation in OASIS KO mice, we compared the gene expression in calvaria between WT and OASIS KO mice using a microarray.
