Expression Data from murine control and IL6ralpha-deficient macrophages stimulated with Interleukin-6

(Submitter supplied) IL-6 induces IL4ralpha expression in macrophages. This mechanism is necessary to promote macrophage polarization towards an M2-phenotype and is crucial to limit the inflammatory response both upon obesity and LPS-endotoxemia. In this dataset, we include the expression data obtained from primary murine bone marrow-derived macrophages from control and IL6ralpha-deficient macrophages (n=4vs4) stimulated with interleukin-6 (IL-6)

Interleukin-6 effect on interleukin-6 receptor α-deficient macrophages

Analysis of interleukin-6 receptor α-deficient bone marrow-derived macrophages (BMDMs) stimulated with interleukin-6 (IL-6). Cytokine IL-6 is linked to obesity, and macrophages are key mediators of obesity-associated inflammation. Results provide insight into the role of IL-6 in macrophages.

IL-6/Stat3-Dependent Induction of Distinct, Obesity-Associated Natural Killer Cells Deteriorates Energy and Glucose Homeostasis

(Submitter supplied) Natural killer (NK) cells contribute to the development of obesity-associated insulin resistance. We demonstrate that in mice obesity promotes the expansion of interleukin-6 receptor (IL6Ra)-expressing NK cells, which also express a number of other myeloid lineage genes such as the colony-stimulating-factor 1 receptor (Csf1r). Selective ablation of Csf1r- expressing NK cells prevents obesity and insulin resistance. more...

Gene expression profile of peroxisome proliferator-activated receptor delta (Ppard)-overexpressing RAW 264.7 macrophages treated vehicle, GW501516 or interleukin-4 (Il-4)

(Submitter supplied) Investigation of whole genome gene expression level changes in GW501516 (a Ppard ligand) or Il-4 treated Ppard-overexpressing RAW 264.7 macrophages as compared to vehicle. Alternative activation of adipose tissue resident macrophages inhibits obesity-induced metabolic inflammation. In the current study we profile genes regulated by two M2 inducers, Il-4 or Ppard agonists and find that alternative activation promotes the cell survival program, while inhibiting that of inflammation-related cell death.

Immunoproteasome dysfunction augments alternative polarization of alveolar macrophages

(Submitter supplied) The proteasome is a central regulatory hub for intracellular signaling by degrading numerous signaling mediators. Immunoproteasomes are specialized types of proteasomes known to be involved in shaping adaptive immune responses, but their role for innate immune signaling is elusive. Here, we analyzed immunoproteasome function for polarization of alveolar macrophages which are highly specialized tissue macrophages of the alveolar surface of the lung. more...

mRNA analysis of gene expression in adipose tissue to compare high fat fed sgp130Fc transgenic vs IL6 knock-out vs wild-type mice

(Submitter supplied) We want to analyse on a broad level the difference of gene expression in the adipose tissue between high fat fed sgp130Fc Transgenic which have no IL6 transsignalling and the IL6 knockout which have no IL6 signalling at all. Because the sgp130Tg mice show a marked adipogenic phenotype we want to compare it with the IL6 KO.

Cx3cl1 expression in the adipose tissue is downregulated during development of obesity

(Submitter supplied) To assess changes in expression level of various chemokines and their receptors on diet-induced obesity, we analysed gene expression in adipose tissue of C56BL/6J mice fed a high-fat (HF) diet or normal chow diet for 8 weeks. HF diet-induced obese (DIO) mice showed adipose tissue inflammation and insulin resistance. Comprehensive gene expression analysis showed that MCP-1–CCR2 and CCL5–CCR5 signalling in epididymal white adipose tissue (eWAT) were enhanced during the development of obesity. more...

LPS hypersensitivity of gp130 mutant mice is independent of elevated hematopoietic TLR4 signaling

(Submitter supplied) The inflammatory mediator IL6 induced by LPS, which signals via TLR4, has been shown to feedback and augment TLR4 signaling when over-produced in LPS hypersensitive gp130F/F mice. The identity of the LPS/TLR4 responsive inflammatory signaling pathways and gene networks which are modulated by IL6 are unknown. Therefore, to understand the molecular consequences of gp130 hyperactivity in non-haemopoietic tissue on LPS-induced systemic inflammation, global gene expression profiling of livers was performed.

Next Generation Sequencing Facilitates Quantitative Analysis of Ern1f/f and Ern1f/fLyz2-Cre macrophage Transcriptomes

(Submitter supplied) Purpose: IRE1 are indicated to play important role in LPS-induced macrophage activation. The goals of this study are to investigate the effects of IRE1a abrogation in macrophages on Interleukin-4-induced alernative activation of macrophages. Methods: Total cellular RNAs of M2-polarized BMDMs by IL-4 treatment were isolated and subjected to commercial RNA-seq analysis. RNA-seq data were uniquely mapped to mm10 reference genome by TopHat v1.4.1. more...
