Nanoemulsion adjuvant W805EC effect on JAWS II dendritic cells: time course

Analysis of JAWS II DCs incubated with nanoemulsion (NE) adjuvant W805EC for up to 24 hrs. W805EC is an oil-in-water emulsion with nanodroplets similar in size to many viruses infecting the nasal mucosa. Results provide insight into the molecular basis of the adjuvant activity of W805EC NE in DCs.

Gene expression data in Bone Marrow Derived Dendritic Cells (BMDC) following nanoemulsion adjuvant exposure

(Submitter supplied) Antigen uptake, processing and presentation by dendritic cells are regulated by complex intra- and inter-cellular signalling events. Typical vaccine adjuvants lead to the transcription of pro-inflammatory cytokines and chemokines which relate to immune induction. We used microarrays to detail the global expression of genes in BMDC underlying emulsion internalization and DC activation with a non-inflammatory nanoemulsion adjuvant.

Next Generation Sequencing Facilitates Quantitative Analysis of Manganese (II) or LPS treated Wild Type BMDC Transcriptomes

(Submitter supplied) Purpose: The goals of this study are to compare the different functions of manganese (II) and LPS on the activation and maturation of antigen presenting cells-BMDCs. Methods: BMDCs were generated by culturing bone marrow cells with 20 ng/ml of IL-4 (Genscript) and 20 ng/ml of GM-CSF (Genscript) at 37°C and 5% CO2 for 7 days. Then BMDCs were treated with MnCl2 (200 μM) or LPS (100 ng/ml) for 20 h. Then mRNAs were purified, sequenced and analyzed. more...

Bisphosphonate effect on murine whole draining lymph nodes before and 8 hours after VSV infection

(Submitter supplied) Bisphosphonates are a class of drugs that are widely used to inhibit loss of bone mass in patients. We show that the administration of clinically relevant doses of bisphosphonates in mice increases antibody responses to live and inactive viruses, proteins, haptens and existing commercial vaccine formulations. Bisphosphonates exert this adjuvant-like activity in the absence of CD4+ and γδ T cells, neutrophils or dendritic cells and their effect does not rely on local macrophage depletion nor does it depend upon Toll-like receptor signaling or the inflammasome. more...
