RRBS investigation of matched side-population trophoblast, cytotrophoblast and extravillous trophoblast populations from 4 human placentae

(Submitter supplied) To better understand how DNA methylation influences placentation, DNA from first trimester primary trophoblast populations (side-population trophoblasts, cytotrophoblasts and extravillous trophoblasts) isolated using FACS underwent reduced representation bisulfite sequencing and were compared to publicly available data of blastocyst-derived and somatic cell populations.

Epithelial-mesenchymal transition in the differentiation of cyto- to extravillous trophoblast

(Submitter supplied) The transformation of cytotrophoblast (CTB) to extravillous trophoblast (EVT) is an essential process for placental implantation. EVT generated at the tips of the anchoring villi migrate away from the placenta and invade the endometrium and maternal spiral arteries, where they modulate maternal immune responses and remodel the arteries into high-volume conduits to facilitate uteroplacental blood flow. more...

Comparison of ITGA2+ trophoblast progenitors to VCT and EVT

(Submitter supplied) During pregnancy, trophoblast cells in the placenta are the only fetal cells in direct contact with the maternal blood and decidua. They have many functions, including transport of nutrients and oxygen, remodelling the uterine arteries, and communicating with maternal cells. Despite their importance in development and in the success of pregnancy, little is known about human trophoblast progenitors and their differentiation. more...

E12.5 CD9+ Mouse Placental Trophoblast Microarray, Wild-type vs c-Met KO

(Submitter supplied) The placenta serves as the structural interface for nutrient and waste exchange for proper fetal development. Although defects in placental function result in various placental disorders, molecular mechanisms orchestrating placental development and function are poorly understood. Gene targeting studies have shown that Hgf or c-Met KO embryos exhibit growth retardation and markedly smaller size of the placenta, and die by E14.5. more...

Derivation of trophoblast stem cells from naïve human pluripotent stem cells [ATAC-seq]

(Submitter supplied) Naïve human pluripotent stem cells (hPSCs) provide a unique experimental platform of cell fate decisions during pre-implantation development, but their lineage potential remains incompletely characterized. As naïve hPSCs share transcriptional and epigenomic signatures with trophoblast cells, it has been proposed that the naïve state may have enhanced predisposition for differentiation along this extraembryonic lineage. more...

TFAP2C regulates transcription in human naive pluripotency by opening enhancers

(Submitter supplied) Naive and primed pluripotent human embryonic stem cells bear transcriptional similarity to pre- and post-implantation epiblast and thus constitute a developmental model for understanding the pluripotent stages in human embryo development. To identify new transcription factors that differentially regulate the unique pluripotent stages, we mapped open chromatin using ATAC-seq and found enrichment of the activator protein-2 (AP2) transcription factor binding motif at naive-specific open chromatin. more...

Gene expression profiles of trophoblast cell lines

(Submitter supplied) Invasive extravillous trophoblasts (EVTs) of the human placenta are critically involved in successful pregnancy outcome since they remodel the uterine spiral arteries to increase blood flow and oxygen delivery to the placenta and the developing fetus. To gain more insights into their biological role different primary cell culture models are commonly utilised. However, access to early placental tissue may be limited and primary trophoblasts rapidly cease proliferation in vitro impairing genetic manipulation. more...

Characterization of primary isolated villous CTB and proximal column EVT from first trimester human placenta.

(Submitter supplied) The goal of this study was to identify the gene expression signatures of two closely related types of trophoblast in human placentas: villous cytotrophoblasts (vCTB) and proximal column extravillous trophoblasts (pcEVT). The two populations were isolated from first trimester placentas and identified using the specific surface markers, EGFR (vCTB) and HLA-G (pcEVT).

Hypoxia directs human villous CTB differentiation towards the EVT lineage in a HIF-dependent manner.

(Submitter supplied) We showed that hypoxia directs first trimester primary villous cytotrophoblast (vCTB) differentiation preferentially towards HLAG+ extravillous trophoblast (EVT). Infection of primary vCTB with ARNT-specific shRNA attenuates this effect, suggesting a role of the intact HIF-complex in hypoxia-directed CTB differentiation into EVT.

Trophectoderm specific function of Atypical Protein Kinase C iota (PKCi) is essential for post-implantation mammalian development.

(Submitter supplied) Atypical protein kinase C iota (Prkci) knockout mice are embryonic lethal at gestation day 9.5 but the underlying molecular mechanisms is not known. Here , using Prkci knockout mouse model , we show that trophectoderm specific function of PKCi is essential for post-implantation mammalian development. We observed that PKCi is expressed predominantly in the trophectoderm lineage and developing placenta region in post-implantation mouse embryos. more...

Comparison of CRISPR-Cas9 genome-edited JEG-3 YAP knockout and wild type choriocarcinoma cells.

(Submitter supplied) The Hippo signalling-dependent co-activator YAP controls organ size by regulating cell proliferation, apoptosis, stem cell renewal and is involved in cell contact inhibition. Aim of this study was to identify human trophoblast-specific YAP regulated gene signatures by comparing YAP wilde type and YAP knock out JEG-3 coriocarcinoma cell lines.
