NF-kB in Tumor Initiation

(Submitter supplied) p65-/-Ras cells show delayed tumor formation in SCID mice. However, after prolonged latency, tumor formation was observed from these mice. To understand the changes of NF-kB regulated genes before and after tumor formation, RNA from p65+/+Ras, p65+/+RasTumor, p65-/-Ras, p65-/-RasTumor cells were isolated and microarray were performed.

NF-κB subunit p65 depletion effect on Ras-induced transformation model: cultured embryonic fibroblasts

Analysis of p65+/+Ras and p65-/-Ras cells generated by infecting NF-κB subunit p65+/+ and p65-/- MEFs with pBabe-HRASG12V. Post-injection of the Ras cells into SCID mice, RasT tumors were removed, reconstituted in culture and examined. Results provide insight into role of NF-κB in tumorigenesis.

The analysis of gene expression profiles of splenocytes from wild type and UBL4A/GdX-/Y mice upon LPS stimulation

(Submitter supplied) Purpose: To compared the gene expression profiles of splenocytes from wild type and UBLA4/GdX (Ubiquitin-like protein 4A, also named GdX) knockout mice upon LPS stimulation. Methods: GdX+/Y and GdX-/Y mice were injected with LPS. Their splenocytes were collected at different time (90min or 6 hrs), and then performed RNA-sequencing analysis. We sequence the samples used BGISEQ-500 platform. Results: The average mapping ratio with reference genome is 93.42%, the average mapping ratio with gene is 83.40%; A total of 17,920 genes were detected.A number of NF-κB targeting genes, such as IL-6, IL-1a and IL-12, have dramatic higher expressions in the splenocytes of GdX+/Y mice than that of GdX-/Y mice. more...

Developmental LTbR synergistically activates TLR4 mediated inflammatory RelA/NF-kB response

In the current study, we identifya critical role of NF-kB system in mediating stimulus specific crosstalk that allows developmental LTbR signals to sustain inflammatory TLR4 induced RelA/NF-kB response and gene expression.
