Early transcriptional signature in murine cDCs post immunisation with VLPs containing TLR ligands

(Submitter supplied) We have investigated the in vivo transcriptional signature in dendritic cells (DCs) from mice immunised with VLPs containing distinct classes of nucleic acid and correlated the expression patterns with the efficiency of induced T cell responses.

Dominant roles of B cells in the initiation of CD4 T cell response to a virus-derived nanoparticle antigen

(Submitter supplied) Nanoparticles, including virus-like particles (VLP), are attractive candidates as carriers for vaccines and drug delivery. However, little is known about how they are targeted to the immune system in vivo. Using RNA phage Qb-derived VLP (Qb-VLP) as a model antigen, we found surprisingly that antigen-specific B cells, instead of dendritic cells (DCs), were the dominant antigen presenting cells that initiate the naïve CD4 T cell activation, and were sufficient to induce T follicular helper cell development in the absence of DCs. more...

Nanoemulsion adjuvant W805EC effect on JAWS II dendritic cells: time course

Analysis of JAWS II DCs incubated with nanoemulsion (NE) adjuvant W805EC for up to 24 hrs. W805EC is an oil-in-water emulsion with nanodroplets similar in size to many viruses infecting the nasal mucosa. Results provide insight into the molecular basis of the adjuvant activity of W805EC NE in DCs.

DALIA-1 Whole Blood Longitudinal Gene Expression

(Submitter supplied) The DALIA-1 trial was designed to evaluate the safety and immunogenicity of a DC-based vaccine generated by culturing blood monocytes with GM-CSF and IFN-a. DCs were pulsed for 24 hrs with the ANRS LIPO5 peptide pool and activated for the last 6 hrs with LPS. In total, 19 asymptomatic HAART-treated patients were included in the study. Microarray samples were collected in both the treatment phase and follow-up phase of the study.

Gene expression analysis of peripheral blood mononuclear cells prior to and following vaccination of human volunteers with the novel TB vaccine candidate MVA85A

(Submitter supplied) A greater understanding of the relationships between a vaccine, the immune response it induces, and protection from disease would greatly facilitate vaccine development. Modified Vaccinia virus Ankara expressing antigen 85A (MVA85A) is a novel tuberculosis (TB) vaccine designed to enhance responses induced by Bacille Calmette-Guerin (BCG). Antigen-specific interferon-γ (IFN-γ) production is greatly enhanced by MVA85A and peaks one week post-vaccination, however, the variability in response between healthy individuals is extensive. more...

Single cell RNA sequencing analysis of CD8+ T cell subsets following DENV-vaccine administration

(Submitter supplied) PBMC-derived CD8 T cell activation and gene expression profiles were assessed 14 days following a single dose of a candidate live-attenuated tetravalent denge virus vaccine (DENVax). Three distinct populations of CD8+ T cells were isolated by flow cytometric sorting: 1) CD3+ CD8+ CD38- HLA-DR- T cells, 2) CD3+ CD8+ CD38+ HLA-DR+ T cells, and 3) CD3+ CD8+ CD71+ HLA-DR+ T cells
