Genome-wide analysis of STAT6 target genes in IL-4 treated human cord blood CD4+ cells

(Submitter supplied) STAT6 is a major transcription factor driving the polarization of Th2 cells in response to cytokine IL-4. Here we have analyzed on a genome wide level the STAT6 mediated gene expression after IL-4 induction in naive human CD4+ T cells. RNAi mediated STAT6 knockdown was used to reveal the genes specifically regulated by STAT6.

Genome-wide detection of STAT6 binding sites in IL-4 treated naive human CD4+ T cells

(Submitter supplied) STAT6 is a major transcription factor driving the polarization of Th2 cells in response to IL-4. STAT6 is phosphorylated by Jak1 and Jak3 kinases at the IL-4 receptor, after which phosphorylated STAT6 forms a homodimer and translocates into the nucleus. There STAT6 binds to specific DNA sequences, regulating the transcription of its target genes. Here we have analyzed on a genome wide level the STAT6 binding sites, after 1h and 4h of IL-4 induction, in naive human CD4+ T cells. more...

Comparative DNA methylation profiling of human monocyte-derived macrophages and dendritic cells

(Submitter supplied) In mammals, the dynamics of DNA methylation, in particular the regulated, active removal of cytosine methylation has remained a mystery, partly due to the lack of appropriate model systems to study DNA demethylation. Previous work largely focused on proliferating cell types that had been mitotically arrested using pharmacological inhibitors to distinguish between active and passive mechanisms of DNA demethylation. more...

Expression analysis of dendritic cells, macrophages and monocytes

(Submitter supplied) Expression profiles at different time points during dendritic cell differentiation (induced by specific culture conditions) including monocytes as well as expression profiles between monocytes and completely differentiated cells (macrophages at day7 and dendritic cells at day7, respectively) were compared. Monocyte-derived dendritic cells (DC) were obtained by culturing elutriated monocytes with 20U/ml IL-4, 280U/ml GM-CSF and 10% FCS; monocyte-derived macrophages (MAC) were obtained by culturing elutriated monocytes with 2% AB serum.

SIGNAL INTEGRATION AND TRANSCRIPTIONAL REGULATION OF THE INFLAMMATORY RESPONSE MEDIATED BY THE GM-/M-CSF SIGNALING AXIS IN HUMAN MONOCYTES [RNA-seq]

(Submitter supplied) In recent years, the M-CSF and GM-CSF cytokines have been identified as opposing regulators of the inflammatory program. However, the two cytokines are simultaneously present in the inflammatory milieu, and it is not clear how cells integrate these signals. In order to understand the regulatory networks associated with the GM/M-CSF signaling axis we analyzed DNA methylation in human monocytes. Our results indicate that GM-CSF induces activation of the inflammatory program and extensive DNA methylation changes, while M-CSF-polarized cells are in a less differentiated state. more...
