Sex-biased expression of small RNA by microarray in critically endangered Amur sturgeon

(Submitter supplied) In this study, we utilized the microfluidics chip technology on the gonads of Amur sturgeon to identifiy gender-specific sRNAs. The probes of all miRNAs about 2751 published in fish and our novel miRNAs from sturgeon were chosed in the microarray experiment.

Small non-coding RNA profiling and the role of piRNA pathway genes in the protection of chicken primordial germ cells

(Submitter supplied) Background: Genes, RNAs, and proteins play important roles during germline development. However, the functions of non-coding RNAs (ncRNAs) on germline development remain unclear in avian species. Recent high-throughput techniques have identified several classes of ncRNAs, including micro RNAs (miRNAs), small-interfering RNAs (siRNAs), and PIWI-interacting RNAs (piRNAs). These ncRNAs are functionally important in the genome, however, the identification and annotation of ncRNAs in a genome is challenging. more...

Developmental regulation of piRNAs during spermatogenesis in Drosophila melanogaster

(Submitter supplied) The piRNA pathway is studied in great detail in Drosophila female germline. In this study we show that unlike the female germline where all Piwi proteins are expressed throughout oogenesis, Ago3 - a Piwi family protein shows a spatial expression male germline. To understand dynamics of piRNA pathway during spermatogonia and primary spermatocyte stages of male germline development, we used arrest mutants. more...

Dynamics of the transcriptional landscape during human gonad development during fetal life

(Submitter supplied) Fetal life is a crucial period for sexual reproduction when bipotential gonads differentiate into either a testis or an ovary. Gaining insights into the complex molecular events underlying this process is central to a better understanding of disorders of sexual development. The present work therefore intends to improve the knowledge on molecular pathways at play during gonad development in humans using RNA-Sequencing.

Vreteno, a gonad-specific protein, is essential for germline development and primary piRNA biogenesis in Drosophila

(Submitter supplied) In Drosophila, Piwi proteins associate with Piwi-interacting RNAs (piRNAs) and protect the germline genome by silencing mobile genetic elements. This defense system acts in germline and gonadal somatic tissue to preserve germline development. Genetic control for these silencing pathways varies greatly between tissues of the gonad. Here, we identified Vreteno (Vret), a novel gonad-specific protein essential for germline development. more...

Vreteno, a gonad-specific protein, is essential for germline development and primary piRNA biogenesis in Drosophila.

(Submitter supplied) Here, we analyzed small RNA libraries derived from ovarian tissues heterozygous or mutant for the Tudor gene, Vreteno. In the absence of vret, Piwi-bound piRNAs are lost without changes in piRNA precursor transcript production, supporting a role for Vret in primary piRNA biogenesis. In the germline, piRNAs can engage in an Aub/Argonaute 3 (AGO3)-dependent amplification in the absence of Vret, suggesting that Vret function can distinguish between primary piRNAs loaded into Piwi/Aub complexes and piRNAs engaged in the amplification cycle. more...

An mRNA/3' UTR-directed primary piRNA pathway in Drosophila ovarian somatic cells

(Submitter supplied) Piwi-interacting RNAs (piRNAs) are ~24-30 nucleotide regulatory RNAs that are abundantly expressed in gonads. The most well-understood piRNAs mediate post-transcriptional defense against transposable elements (TEs), and derive from sense or antisense strands as a consequence of "ping-pong" amplification of complementary sequences of active TEs and piRNA master control transcripts. Another class of piRNAs, such as those expressed in pachytene testis, derive from large intergenic clusters that are strictly single-stranded. more...

The porcine piRNA transcriptome response to Senecavirus A infection

(Submitter supplied) Senecavirus A (SVA) belongs to the family of small RNA viruses, the genus Senecavirus, and has become a research hotspot because of the oncolytic viral characteristics. PIWI-interacting RNAs (piRNAs) are a class of small RNAs found in mammalian cells in recent years; however, the host piRNA expression profile during SVA infection and their role in viral infection is not well understood. In this study, we obtained small RNA transcriptome expression profiles from SVA-infected pig kidney cell lines (PK-15) by high-throughput sequencing. more...

Identification and characterization of microRNAs and Piwi-interacting RNAs in cockerels’ sperm by Solexa sequencing

(Submitter supplied) There have been substantial researches focused on the roles of microRNAs (miRNA) and Piwi-interacting RNAs (piRNA) delivered from mammalian spermatozoa, comparatively little is known about the role of sperm-delivered miRNAs and piRNAs within cockerels’ (breeding cock) sperm. A small RNA library of cockerels’ sperm was constructed using Illumine high-throughput sequencing technology.

FLX sequencing of Piwi-associated small RNAs extracted from Drosophila ovarian somatic cells (OSC)

(Submitter supplied) piRNAs function in silencing retrotransposons by associating with the PIWI proteins, AGO3, Aub, and Piwi, in Drosophila germlines. Bioinformatics analyses of piRNAs in Drosophila ovaries suggested that piRNAs are produced by two systems, the primary processing pathway and the amplification loop, from repetitive genes and piRNA clusters in the genome. The amplification loop occurs in a Dicer-independent, PIWI-Slicer-dependent manner. more...

Small RNA and gene expression profile in the adult testes of the common marmoset

(Submitter supplied) Small RNAs mediate gene silencing by binding Argonaute/Piwi proteins to regulate target RNAs. Here we describe small RNA profiling of the adult testes of Callithrix jacchus, the common marmoset. The most abundant class of small RNAs in the adult testis was piRNAs, while 353 novel miRNAs but few endo-siRNAs were also identified. MARWI, a marmoset homolog of mouse MIWI and a very abundant PIWI in adult testes, associates with piRNAs that show characteristics of mouse pachytene piRNAs. more...
