Circular RNAs in cattle muscle: developmental stage-dependent changes

(Submitter supplied) Circular RNAs (circRNAs) have been identified in various tissues and cell types from human, monkey, porcine and mouse. However, expression profile of circRNAs across cattle muscle development has never been investigated. Here, we examine the expression of circRNA in cattle skeletal muscle at embryonic stage and adult stage, exhibiting the first report of circRNA in the cattle muscle development of a large animal. more...

RNA-seq of RNase R treated and ribo- RNAs from leg muscle of Xinghua Chickens

(Submitter supplied) We have used RNA-seq to examine circular RNAs from RNase R treated and ribo- RNAs in chicken leg muscle of three different development stages (11 embryo age, 16 embryo age, and 1 days post hatch). Our study reveals the prevalence of circRNAs in chicken, and has identified circRNAs differentially abundant in different stage of embryonic skeletal muscle, suggesting its important functions during poultry muscle development.

Guanidineacetic acid regulate myogenic differentiation through miR-133a-5 and miR-1a-3p co-mediated PI3K-Akt-mTOR signaling pathway

(Submitter supplied) Through small RNA sequencing, we finded that a total of 8 miRNAs, including miR-133a-3p and miR-1a-3p cluster, showed differential expression after guanidineacetic acid supplement. To further study the function of miR-133a-3p and miR-1a-3p in guanidineacetic acid induce myotube hypertrophy, we transfected miR-133a-3p and miR-1a-3p mimics, that also induce myotube hypertrophy. Through bioinformatics and dual-luciferase reporter system, the target gene of miR-133a-3p and miR-1a-3p were respectively determined. more...

RNA-sequencing of chicken primary myoblasts and myotubes

(Submitter supplied) Chick is an ideal model system for myogenesis study. Lots of genes would change their expression during myogenesis, however, is there any difference about gene expression between chick and mice? Our study represents the first detailed analysis of the transcriptomes of chicken primary myoblasts and myotubes, with biologic replicates, generated by RNA-seq technology. The data reported here may provide an important understanding of the genes involved in the regulation of chicken myogenesis.

Genome-wide maps of c-Myc binding sites in myoblasts and myotubes

(Submitter supplied) By applying ChIP-seq, we generated genome-wide maps of c-Myc in skeletal myoblasts and myotubes. We found that c-Myc binds to 19354 confident target with a large portion residing in the intergenic regions. In addition, c-Myc was found to regulate many loci. Further detailed study revealed that c-Myc can regulate some genes, miRNAs, and lincRNAs which are fucntional in skeletal myogenesis. In this study, we identified a c-Myc induced regulatory pathway in myogensis.

miR-487b, miR-3963 and miR-6412 delayed myogenic differentiation in C2C12 mouse myoblast-derived cells.

(Submitter supplied) We newly identified skeletal muscle differentiation-associated miRNAs by comparing miRNA expression profile between C2C12 cell and Wnt4-overexpressing C2C12 cell. miR-487b, miR-3963 and miR-6412 are significantly down-regulated in differentiating C2C12 cells, and transfection of their mimics resulted in reduced expression of myogenic differentiation markers including Troponin T, myosin heavy chain fast and slow type.

Transcriptomics analysis of proliferation and differentiation of Guangxi cattle muscle stem cells

(Submitter supplied) More and more studies have shown that mRNAs and non-coding RNAs are involved in human diseases and various biological processes of animals, and play an important functional role in animal muscle growth and development. In this study, the expression levels of myogenic marker molecules MYOD1, MYOG, and MyHC increased after cattle muscle stem cells transition from proliferation to differentiation, which promoted the differentiation and maturation of cells. more...
