Profiles of pure CD4+ and CD8+ T cells subsets from HIV-infected nonprogressors who controlled viremia were indistinguishable from HIV-uninfected individuals. Memory-like CD8 T-cells sustain the immune response to chronic viral infections.

(Submitter supplied) We have discovered a small subpopulation of virus-specific CD8 T-cells that sustains the T-cell response in chronic infections. These cells are defined by - and depend on - the expression of the transcription factor Tcf1 (T cell factor 1) and show key characteristics of central memory cells while lacking an effector signature. Unlike conventional memory cells, Tcf1+ T-cells display hallmarks of an “exhausted” phenotype, including the expression of certain inhibitory receptors.

Mechanisms of Restoring T Cell Immunity after Cure of Chronic Viral Infection

(Submitter supplied) Understanding the molecular mechanisms of T cell exhaustion and reinvigoration is crucial to improving T cell based immunotherapy. In this study we confirmed key differences between memory and exhausted antigen-specific CD8+ T cells in hepatitis C virus infection before and after antiviral treatment. After viral cure, we observed the although phenotypically they seem to recover, functionally they showed little improvement and critical transcriptional regulators remained in exhausted state.
