Gene co-expression networks drive and predict reproductive effects in Daphnia in response to environmental disturbances

(Submitter supplied) ncreasing effects of anthropogenic stressors and those of natural origin on aquatic ecosystems have intensified the need for predictive and functional models of their effects. Here, we use gene expression patterns in combination with weighted gene co-expression networks and generalized additive models to predict effects on reproduction in the aquatic microcrustacean Daphnia. We developed models to predict effects on reproduction upon exposure to different cyanobacteria, different insecticides and binary mixtures of cyanobacteria and insecticides. more...

Assessment of chemical mixtures and ground water effects on Daphnia magna transcriptomics

(Submitter supplied) Small organisms can be used as biomonitoring tools to assess chemicals in the environment. Chemical stressors are especially hard to assess and monitor when present as complex mixtures. Here, Daphnia magna were exposed for 24 hours to five different munitions constituents 2,4,6-trinitrotoluene (TNT), 2,4-dinitrotoluene (2,4-DNT), 2,6-dinitrotoluene (2,6-DNT), trinitrobenzene (TNB), dinitrobenzene (DNB), or 1,3,5-trinitro-1,3,5-triazacyclohexane (RDX) as well as to 8 different munitions mixtures and ground water contaminated with munitions constituents. more...

Gene expression profiling in Daphnia magna, Part III: Uncovering Biomarkers for Metal and ORC exposures

(Submitter supplied) To determine toxicant specific effects of Ordnance Related Compound (ORC) exposure we performed microarray hybridizations with RNA isolated from Daphnia magna following different ORC exposures at the 1/10 LC50. The gene expression profiles revealed toxicant specific gene expression profiles allowed for the identification of specific biomarkers of exposure. Keywords: ecotoxicogenomic exposure study

Nickel and binary metal mixtures in Daphnia magna after 96h

(Submitter supplied) The recent development of a custom cDNA microarray platform for one of thé standard organisms in aquatic toxicology, Daphnia magna, opened up new ways to mechanistic insights of toxicological responses. In this study, gene expression and (sub)organismal responses (Cellular Energy Allocation, growth) were assayed after short-term waterborne metal exposure. Microarray analysis of Ni-exposed daphnids revealed several affected functional gene classes, of which the largest ones were involved in different metabolic processes (mainly protein and chitin related processes), cuticula turnover, transport and signal transduction. more...

Transgenerational inheritance of DNA hypomethylation in Daphnia magna in response to salinity stress.

(Submitter supplied) Epigenetic mechanisms have been found to play important roles in environmental stress response and regulation. These can, theoretically, be transmitted to future unexposed generations, yet few studies have shown persisting stress-induced transgenerational effects, particularly in invertebrates. Here, we focus on the aquatic microcrustacean Daphnia, a parthenogenetic model species, and its response to salinity stress. more...

Effects of Single and Combined Low Concentrations of Neuroactive Drugs on Daphnia magna Reproduction and Transcriptomic Responses

(Submitter supplied) Transcriptomic profiling of Daphnia magna samples exposed to carbamazepine (CBZ), diazepan (DZP), propranolol (PR) and to an equitoxic mixture of them (M). Assessing the risks of emerging contaminants such as neuro-active pharmaceuticals in the environment requires an understanding of their joint effects at low concentrations across species. Here, we assessed reproductive and transcriptional effect of propranolol, diazepam and carbamazepine on the crustacean Daphnia magna at environmentally relevant concentrations. more...
