Transcriptome expression study for miRNA-mRNA target discovery

(Submitter supplied) Total RNA was collected from DU145-control(scr) and DU145-miR-106a transfected cells. Gene expression analysis was performed by The Centre for Applied Genomics (The Hospital for Sick Children, Toronto, Canada) using Affymetrix GeneChip Human Gene 2.0 ST array. Data were normalized using the default parameters in Affymetrix Expression Console Software 1.4. Genes downregulated 0.8-fold in miR-106a compared to control(scr) were identified as possible mRNA targets.

Oxidative stress-mediated alterations of the microRNA expression profile in porcine granulosa cells using RNA-Seq technique

(Submitter supplied) To investigate the effects of oxidative stress induced by hydrogen peroxide(H2O2) on miRNA expression in porcine granulosa cells,we examined the impact of H2O2-mediated oxidative stress in procine GC through miRNA-Seq. We identified 33 (19 upregulated, 14downregulated) and 22 (14 upregulated, 8 downregulated miRNAs) differentially expressed miRNAs (DEmiRNAs) at 300μM and 100μM H2O2, respectively, compared with the control group.

Differentially expressed miRNA in biological premature ovarian insufficiency (bPOI)

(Submitter supplied) We first examined changes of miRNAs and expression using microarrays ( Exiqon miRCURY™ microarray LNA v.18.0) of GCs from 10 bPOI and 10 age and BMI matched normal control women. Seventy-five miRNAs (Fold Change [FC]>2, P<0.05) were identified differentially expressed between bPOI patients and control women.
