RNA sequencing data of aging zebrafish brain progenitor cells

(Submitter supplied) Our aim in this study was to investigate the gene expression changes through pre-adult to adult, and adult to old age, in zebrafish brains. We isolated the neuronal cells and performed RNA sequencing in 3 months old, 6-8.5 months old, and 26-31 months old zebrafish brains. The cells were enriched in progenitor cell markers, which diminished throughout the aging process.

Gene expression assay for ALCAM depleted endometrial cancer cell lines

(Submitter supplied) Endometrial cancer is the most common gynaecological cancer in western countries, being the endometrioid tumours the most common subtype. Most patients are diagnosed at an early stage and present an excellent prognosis. However, a consistent number of those continue to suffer recurrence, without means to be identified by risk classification systems. Thus, finding a reliable marker to predict recurrence becomes an important unmet clinical issue. more...

Expression data from hsa-mir-125 transfected MCF7 cells

(Submitter supplied) MicroRNA (miRNA) deregulation has long been associated with various cancers. Among an expanding list of cancer related miRNAs, deregulation of miR-125b has been well documented in various cancers including breast. Based on current knowledge, miR-125b is considered to be a tumor suppressor in breast cancers. While important mRNA targets have been defined for miR-125b, here, we aimed to further investigate direct/indirect consequences of miR-125b expression in breast cancer cells by using a transcriptome approach. more...

Interleukin-4 induces neural stem cell plasticity by antagonizing Serotonin/HTR1 signaling that suppresses BDNF/NGFRA in adult zebrafish Alzheimer's disease model

(Submitter supplied) In Alzheimer’s disease (AD), reduced neural stem cell (NSC) plasticity causes reduced neurogenesis. Therefore, supplying the brain with new neurons using endogenous neurogenic reservoir – NSCs - might counteract the progression of AD. However, the mechanisms that enhance NSC plasticity are unknown. We recently generated an AD model in adult zebrafish brain where NSCs could react by enhanced plasticity and neurogenesis, and identified Interleukin-4 (IL4) as a key factor underlying this ability. more...

Single cell analyses of Amyloid-beta42 and Interleukin-4 on neural stem cell plasticity I

(Submitter supplied) The neural stem cell (NSC) reservoir can be harnessed for stem cell-based regenerative therapies. Zebrafish remarkably regenerate their brain by inducing NSC plasticity in a Amyloid-β-42 (Aβ42)-induced experimental Alzheimer’s disease (AD) model. Interleukin-4 (IL-4) is also critical for AD-induced NSC proliferation. However, the mechanisms of this response have remained unknown. Using single-cell transcriptomics in the adult zebrafish brain, we identify distinct subtypes of NSCs and neurons and differentially regulated pathways and their gene ontologies and investigate how cell-cell communication is altered through ligand-receptor pairs in AD conditions. more...

Transcriptomic profiling of microglia and astrocytes throughout aging

(Submitter supplied) Purpose:The goal of this study are to understand the role of miceoglia and astrocyte in both physiological and pathological conditions. Methods: Microglia and astrocyte were isolated from 5 time points WT/APP-PS1 mice in triplicate. mRNA were generated by SMART seq2 using Illumina Novaseq platform. Conclusions: Using DESeq2 to sequence polyA-selected mRNAs from microglia and astrocytes isolated from whole brain. more...

Gene expression in primate postnatal brain through lifespan - cerebellar cortex

(Submitter supplied) We investigated molecular changes during human, chimpanzee, and rhesus macaque postnatal brain development at the transcriptome, proteome, and metabolome levels in two brain regions: the prefrontal cortex (PFC) that is involved in several human-specific cognitive processes, and the cerebellar cortex (CBC) that may be functionally more conserved. We find a nearly three-fold excess of human-specific gene expression changes in PFC compared to CBC. more...

miRNA expression data from rhesus macaque postnatal brain in prefrontal cortex: time course

(Submitter supplied) Gene expression changes determine functional differentiation during development and are associated with functional decline during aging. While developmental changes are tightly regulated, regulation of aging changes is not well established. To assess the regulatory basis of age-related changes and investigate the mechanism of regulatory transition between development and aging, we measured mRNA and microRNA expression patterns in brains of humans and rhesus macaques over the entire species’ lifespan. more...

Alterations of immune cells heterogeneities in the aged zebrafish brain at single-cell resolution II

(Submitter supplied) Immune cells, the most core part of the immune system of the whole body, could rapidly response to the change of environment. Its roles in tissue homeostasis and disease were fairly clear except brain, an immune privilege site. Previous studies had revealed the contribution of microglia, a tissue-resident macrophage, for brain development and neuronal disease. But it was currently unknown whether the other subtypes of immune cells and which of the microglia populations program their function during different stages of development. more...
