Transforming growth factor beta 1 induces methylation changes in lung fibroblasts [Affymetrix]

(Submitter supplied) Idiopathic pulmonary fibrosis (IPF) is a complex disease of unknown etiology. Environmental factors can affect disease susceptibility via epigenetic effects. Few studies explore global DNA methylation in lung fibroblasts, but none have focused on transforming growth factor beta-1 (TGF-b1) as a potential modifier of the DNA methylome. Here we analyzed changes in methylation and gene transcription in normal and IPF fibroblasts following TGF-b1 treatment.

Global DNA Methylation Levels in Lung Fibroblasts from Patients with Idiopathic Pulmonary Fibrosis Compared to Nonfibrotic Controls

(Submitter supplied) Fibroblasts are the key effector cells in idiopathic pulmonary fibrosis (IPF), a chronic, progressive scarring disorder that results in impaired gas exchange and respiratory failure. Fibroblasts from IPF patients exhibit aberrant expression of multiple genes, but the DNA methylome of IPF fibroblasts has never been characterized. We utilized the HumanMethylation27 array, which assays the DNA methylation level of 27,568 CpG sites to compare the DNA methylation patterns of IPF fibroblasts (n=6) with those of nonfibrotic patient controls (n=3) and commercially available normal lung fibroblast cell lines (CCL190, CCL204, and CCL210). more...

Forkhead Box F1 (FOXF1) represses fibroblast functions relevant to fibrogenesis

(Submitter supplied) Aberrant expression of master phenotype regulators by lung fibroblasts may play a central role in idiopathic pulmonary fibrosis (IPF). Interrogating IPF fibroblast transcriptome datasets, we identified Forkhead Box F1 (FOXF1), a DNA-binding protein required for lung development, as a candidate actor in IPF. Thus, we determined FOXF1 expression levels in fibroblasts cultured from normal or IPF lungs in vitro, and explored FOXF1 functions in these cells using transient and stable loss-of-function and gain-of-function models. more...

Global Methylation Patterns in Idiopathic Pulmonary Fibrosis

(Submitter supplied) Idiopathic Pulmonary Fibrosis (IPF) is a chronic progressive lung disease that affects more than 5 million people worldwide with a steady increase in both incidence and mortality. There is currently no effective therapy and the median survival without transplant is 2-5 years. The etiological factor is unknown, but several observational and pathogenesis studies suggest that environmental agents may cause IPF. more...

Transcriptomic analysis of miR-19a-19b-20a subcluster-overexpressed fibroblasts in bleomycin-induced lung fibrosis

(Submitter supplied) Lung fibroblasts play a pivotal role in pulmonary fibrosis, a devastating lung diseases, by producing extracellular matrix. MicroRNAs (miRNAs) suppress a lot of genes posttranscriptionally, but the dynamics and the role of miRNAs in activated lung fibroblasts in fibrotic lung has been poorly understood. We found miR-19a, 19b and 20a subcluster expression increased in activated lung fibroblasts as the fibrosis progression. more...

Time-course global miRNA expression profiling on lung fibroblasts isolated from untreated, bleomycin-treated and silica-treated mice

MicroRNAs (miRNAs) suppress a lot of genes posttranscriptionally, but the dynamics and the role of miRNAs in activated lung fibroblasts in fibrotic lung has been poorly understood. To elucidate these problems, we performed global miRNA expression profiling of lung fibroblasts of bleomycin- and silica-induced fibrotic lungs

Aberrant DNA methylation in Non Small Cell Lung Cancer associated fibroblasts

(Submitter supplied) Epigenetic changes through altered DNA methylation have been implicated with critical aspects of tumor progression, and have been extensively studied in cancer cells and tumor tissue samples. In contrast, our current knowledge of the aberrant genomic methylation in tumor-associated fibroblasts (TAFs), which are critical co-conspirators of tumor progression, is very scarce. To address this gap of knowledge, we conducted genome-wide DNA methylation profiling on lung TAFs and paired control fibroblasts (CFs) from surgical non-small cell lung cancer patients. more...

Identification of the gene expression in IPF lung fibroblasts after demethylation

(Submitter supplied) Idiopathic pulmonary fibrosis (IPF) is a progressive lung disease. Although the pathogenesis is poorly understood, evidence suggests that genetic and epigenetic alterations, such as DNA methylation, may play a key role. We used microarrays to see the gene expression in IPF fibroblasts after demethylation agent 5'-azacytidine treatment.
