Comparison of testis transcriptome changes resulting from mutations in dany and representative meiotic arrest genes

(Submitter supplied) The Drosophila gene dany is expressed specifically in spermatocytes and encodes a nuclear protein with similarity to the products of the genes distal antenna (dan) and distal antenna-related (danr). Loss of dany function results in male sterility. Mutant spermatocytes fail to differentiate similar as previously observed in mutants lacking tTAF and tMAC function. Microarray analyses were performed to characterize the effects of loss of dany function on gene expression in testis and for a comparison of the effects with those caused by mutations in the tTAF gene spermatocyte arrest (sa) and the tMAC genes always early (aly) and matotopetli (topi)

Screening of genes involved in chromosome segregation in meiosis I

(Submitter supplied) Prophase I of meiosis involves dynamic chromosome segregation processes including synapsis, meiotic recombination, and cohesion. Genetic defects in genes participating in these processes consistently cause reproduction failure in mice. To identify candidate genes responsible for infertility or recurrent pregnancy loss in humans, we performed expression profiling of male and female gonads of mice undergoing meiotic prophase I.

Gonadal development: ovaries and testes

Analysis of gonads from fetal, neonatal and adult C57BL/6 males and females. Events during meiosis I prophase are critical for correct segregation of chromosomes. Defects in these events may contribute to infertility. Results provide insight into molecular mechanisms underlying meiosis I prophase.

Principles of 3D chromosome folding and evolutionary genome reshuffling in mammals

(Submitter supplied) Here we describe the principles of 3D genome folding dynamics in vertebrates and show how lineage-specific patterns of genome reshuffling can result in different chromatin configurations. We (i) identified different patterns of chromosome folding across vertebrate species, (ii) reconstructed ancestral marsupial and afrotherian genomes analyzing whole-genome sequences of 10 species representative of the major therian phylogroups, (iii) detected lineage-specific chromosome rearrangements and (iv) identified the dynamics of the structural properties of genome reshuffling through therian evolution.
