Epidermal growth factor effect on HER2-overexpressing mammary epithelial MCF10A cells: time course

Analysis of MCF10A and HER2-overexpressing MCF10A cells grown on Matrigel in the absence or presence of epidermal growth factor (EGF). MCF10A-HER2 cells exhibited autonomous growth, independent of growth factors. Results provide insight into molecular bases underlying the growth autonomy confer

A Splice Variant of HER2 Activates Key Signaling Cascades and Evokes Mammary Tumors and Metastases

(Submitter supplied) The Epidermal Growth Factor Receptor 2 (ERBB2 or HER2) is amplified and overexpressed in approximately 20% of invasive breast cancers and is associated with metastasis and poor prognosis. Here we describe the role of a constitutively active splice variant of HER2 (Delta-HER2) in human mammary epithelial cells. Overexpression of Delta-HER2 in human mammary cells decreased apoptosis and increased proliferation and expression of epithelial-to-mesenchymal markers. more...

Effects of acute Notch activation on the mammary epithelial compartment in vivo

(Submitter supplied) Notch signaling is widely implicated in mouse mammary gland development and tumorigenesis. To investigate the effects of acute activation of Notch signaling in the mammary epithelial compartment, we generated bi-transgenic MMTV-rtTA; TetO-NICD1 (MTB/TICNX) mice that conditionally express a constitutively active NOTCH1 intracellular domain (NICD1) construct in the mammary epithelium upon doxycycline administration.

Ductal Carcinoma Progression

(Submitter supplied) To obtain insight into the molecular basis of ductal carcinoma in situ (DCIS) and its progression by infiltrating surrounding tissue, we have performed cellular-based gene expression analysis of pure DCIS and DCIS with co-existing invasive ductal carcinoma (IDC) and compared the histological and molecular aspects between these morphologically identical lesions seeking to find key genes involved in DCIS progression. more...

Transcriptome profiles of mouse mammary stem cells and progenitor cells

(Submitter supplied) MaSC, Luminal progenitor enriched subpopulations were isolated from virgin and pregnant mice based on using both surface marker or internal reporter transgene (GFP) expression. (Tiede BJ et al., 2009, PLoS ONE 4(11): e8035. doi:10.1371/journal.pone.0008035), and the transcirptome profiles were determined and compared.

Chimeric EGFR ChIP-seq of an MCF-10A clone expressing chimeric EGFR and Erbb2 receptors

(Submitter supplied) Synthetic activation of chimeric EGFR-Erbb2 receptors causes nuclear translocation of the heterodimers at long time scales. To identify genomic loci bound by nuclear EGFR-Erbb2, chromatin from an MCF10A-5E clone coexpressing chimeric EGFR and Erbb2 was prepared with or without synthetic activation and immunoprecipitated for chimeric EGFR by its GluGlu epitope tag.

Stochastic 10-cell profiling of an MCF-10A clone expressing chimeric EGFR and Erbb2 receptors

(Submitter supplied) ErbB2 activation of MCF10A cells gives rise to a multiacinar phenotype that is incompletely penetrant. To identify candidate upstream regulators of the phenotype, an MCF10A-5E clone was subcloned to express chimeric receptors of human EGFR and rat Erbb2, and this B2B1 subclone was stochastically profiled (PMID: 20228812) after 24 hours of 3D culture with or without synthetic dimerization.

DEEP SEQUENCING OF MODELS OF BREAST DUCTAL CARCINOMA IN SITU REVEALS ALDH5A1 AS A NOVEL POTENTIAL THERAPEUTIC TARGET

(Submitter supplied) We attempted to identify alterations in gene expression that occur during the progression from normal breast to ductal carcinoma in situ (DCIS) with the aim to elucidate significant genes and pathways underlying the premalignant transformation. To determine the expression changes that are common to multiple DCIS models (MCF10.DCIS, SUM102 and SUM225) and normal mammary epithelial cells (MCF10A), we grew the cells in three dimensional overlay culture with reconstituted basement membrane and used the extracted RNA for 76 cycles of deep sequencing (mRNA-Seq) using Illumina Genome Analyzer GAIIx. more...

Outer ECM-attached vs. inner cells of MCF10A acini at day 6 of morphogenesis

(Submitter supplied) Basal-like carcinoma is a subtype of breast cancer that is characterized by poor prognosis and high intratumor heterogeneity. Using a basal-like breast epithelial line, we have identified two anti-correlated gene-expression programs that arise among single extracellular matrix (ECM)-attached cells during organotypic 3D culture. The first program contains TGFBR3, a high-affinity receptor for transforming growth factor β (TGFβ) and other related ligands. more...
