Lung epithelium response to fungal allergen Alternaria

Analysis of bronchial brushings of lung epithelium from C57BL/6 females after Alternaria challenge. Alternaria allergen is associated with severe asthma, including life-threatening exacerbations. Results provide insight into the molecular basis of the acute innate airway response to Alternaria.

Expression data from mouse lung epithelium after allergen challenge

(Submitter supplied) Changes occur in the lung epithelium after allergen challenge that may give insight into asthma pathogenesis and we sought to identify novelepithelial genes induced by allergen exposure. We used microarrays to identify up-regulated genes induced by allergen challenge in lung epithelium.

Expression data from wild type or FIZZ2 knockout murine lungs

(Submitter supplied) To study the possible fibrotic role of FIZZ2, bleomycin was used to induce pulmonary fibrosis in wild type and FIZZZ2 knockout mice, lungs were then harvested and processed for RNA isolation. We used microarrays to detail the global gene expression regulated by FIZZ2 during fibrotic process.

Gene expression in alveolar macrophages between bleomycin–treated wild-type and S1pr2-/- mice.

(Submitter supplied) We compared mRNA expression in alveolar macrophages between bleomycin–treated wild-type and S1pr2-/- mice, using DNA microarray analysis. In S1pr2-/- macrophages, 398 genes showed decreases to less than 50% of the levels in wild-type macrophages. In contrast, 122 genes showed more than 2.0-fold increases in S1pr2-/- macrophages compared with wild-type macrophages. The downregulated genes in S1pr2-/- mice included the following potentially fibrosis–related genes: profibrotic cytokines, chemokines, and the markers characteristic of classically activated (M1) and alternatively activated (M2) macrophages.

Unconventional ST2- and CD127-negative lung ILC2 populations are induced by Alternaria

(Submitter supplied) Background: Alternaria exposure is associated with severe asthma in humans. Alternaria exposure in mice potently activates group 2 innate lymphoid cells (ILC2s) via the IL-33/ST2 axis and causes ILC2s to robustly secrete type 2 cytokines.  Objective: Our aim was to determine whether conventionally used ILC2 markers, ST2 (IL-33R) and CD127 (IL-7Ra), were sufficient to identify all Th2-cytokine producing ILCs after Alternaria exposure. more...

To Study the effects of antioxidant on allergic airways inflammations

(Submitter supplied) Ragweed challenge in Ragweed (RWE) sensitized animals generates Reactive oxygen species (ROS) in the airway epithelium and induces allergic airway inflammation. We want to study the genes induced by ROS generated by RWE. This goal can be achieved by comparing PBS challenge vs. RWE challenge. Keywords: Infection

STAT6 Blockade Abrogates Aspergillus-Induced Eosinophilic Chronic Rhinosinusitis and Asthma, a Model of Unified Airway Disease

(Submitter supplied) Unified airway disease, including concurrent asthma and chronic rhinosinusitis (CRS), is a common, but poorly understood disorder with no curative treatment options. To establish a murine model of chronic unified eosinophilic airway inflammation, mice were challenged with Aspergillus niger, and sinonasal mucosa and lung tissue were evaluated by immunohistochemistry, flow cytometry and gene expression. more...

Human bronchial epithelial cells exposed to A. alternata spores

(Submitter supplied) This present study is the first to investigate the global changes in host gene expression during the interaction of human bronchial epithelial cells and live Alternaria spores. Human bronchial epithelial cells (BEAS2-B) were exposed to spores or media alone for 24 hours. RNA was collected from three biological replicates/treatment and used to assess changes in gene expression patterns using Affymetrix Human Genome U133 Plus 2.0 Arrays. more...
