Transcription factor Nrf2 gain-of-function model: postnatal day 2.5 whole skin

Analysis of whole skin from female, K5cre-caNrf2 transgenics at P2.5. Nrf2 activation induced epidermal thickening and hyperkeratosis resembling human skin disease lamellar ichthyosis. Results provide insight into the role of Nrf2 as a regulator of protective functions of the epidermis.

Functional analysis of the cytoprotective transcription factor Nrf2 in skin morphogenesis and disease: Identification of Nrf2 target genes

(Submitter supplied) The Nrf2 transcription factor is a key player in the cellular stress response, which regulates the expression of important antioxidant enzymes and other cytoprotective proteins. We recently generated a novel transgenic mouse model to determine the function of Nrf2 in the skin. These mice revealed interesting phenotypic abnormalities, including hyperkeratosis and acanthosis. To gain insight into the underlying molecular mechanisms, we wanted to identify genes, which are differentially expressed in the skin of wild-type and mutant mice before the onset of phenotypic abnormalities.

Gene expression analysis of human keratinocytes (HaCaTs) after non-thermal plasma treatment

(Submitter supplied) Investigation of gene expression in human skin keratinocytes (HaCaT) following non-thermal plasma treatment for 20 s, 60 s, and 180 s compared to untreated and H2O2-treated controls. Microarrays were used to analyze and investigate the biological effects of non-thermal plasma on human keratinocyte cells. Using an argon plasma jet kinpen, regulated transcripts were analyzed and further described in Schmidt et al. more...

Keratinocyte-Macrophage Crosstalk by the Nrf2/Ccl2/EGF Signaling Axis Orchestrates Tissue Regeneration

(Submitter supplied) Unveiling the molecular mechanisms underlying tissue regeneration provides new opportunities to develop treatments for diabetic ulcers and other chronic skin lesions. Here, to gain insight into critical wound response mechanisms regulated by epidermal Nrf2, we micro dissected a ~2.5mm concentric ring around the 10mm wound at 5 days-post-wounding (DPW) and used flow cytometry to isolate wound-associated keratinocytes from Nrf2 +/+ Ker and Nrf2 ∆/∆ Ker mice for bulk RNA-seq. more...

Small proline-rich proteins 1 and 2 function as antimicrobial proteins in the skin
