Rheumatoid arthritic synovium response to rituximab

Analysis of synovial biopsy samples from anti-TNF resistant rheumatoid arthritis (RA) patients 12 wks after the start of rituximab (RTX) therapy. RTX is a mAb against CD20, expressed on subsets of B cells. Results provide insight into how RTX-induced B cell depletion interferes with RA pathogenesis.

Differential effects of TNFalpha and IL1beta on FLS global gene expression profile

(Submitter supplied) TNFalpha and IL1beta play a pathogenic role in rheumatoid arthritis. Both cytokines are known to activate cytokine and metalloproteinase secretion by synovial fibroblasts. In the present study, we wanted to investigate whether TNFalpha and IL1beta displayed differential effects on cultured Fibroblast-like Synovial Cells derived from RA patients. Global gene expression analyses indicated that both cytokines induced similar genes in these cells.

Comparison of gene expression profiles in paired synovial biopsies from rheumatoid arthritis (RA) patients

(Submitter supplied) We explored transcriptomic profiles of paired synovial biopsies from RA patients, in order to assess heterogeneity in synovial tissue at the individual level. Pairwise comparison of transcriptomic profiles showed similar expression of RA-related molecular pathways (TCR signaling, T cell co-stimulation and response to TNF⍺), thereby showing that global molecular alterations in RA synovitis are similar between small and large joints from the same patient.
