PI3K/mTOR Inhibitor NVP-BEZ235 and taxotere effects on prostate cancer xenograft tumors

Analysis of DU145 xenograft tumors in NOD/SCID mice treated with NVP-BEZ235, taxotere, or both. NVP-BEZ235 targets prostate cancer progenitor cells. Taxotere targets the bulk tumor. Results provide insight into potential biomarkers associated with cancer stem cells in these xenograft tumors.

Gene expression analysis of in vivo-grown tumors treated with compounds that either de-bulk the tumor or target cancer stem cells

(Submitter supplied) A large body of evidence has demonstrated that many human tumors are maintained by a small cell population called cancer stem cells (CSCs) or tumor progenitors, which are responsible for tumor formation, therapy resistance and metastasis. We found that ionizing radiation treatment enriches for the CSC phenotype and properties by preferential survival and expansion of tumor progenitor cells. Our studies revealed that aldehyde dehydrogenase (ALDH) activity is indicative of prostate tumor progenitor cells with increased chemo- and radioresistance, enhanced migratory potential, improved DNA- double strand break repair and activation of the signaling pathways, which promote self-renewal and epithelial-mesenchymal transition. more...

Genomic characterization of ovarian cancer spheroids

(Submitter supplied) Spheroids are 3D multi-cell aggregates formed in non-addherent culture conditions. In ovarian cancer (OC), they serve as a vehicle for cancer cell dissemination in the peritoneal cavity. We investigated genes and networks upregulated in three dimensional (3D) versus two-dimensional (2D) culture conditions by Affymetrix gene expression profiling and identified ALDH1A1, a cancer stem cell marker as being upregulated in OC spheroids. more...

Comparative analysis of gene expression profiles comparing radioresistant to radiosensitive prostate cancer cell lines

(Submitter supplied) Prostate cancer cell lines DU145 and LNCaP were purchased from the American Type Culture Collection. Radioresistant (RR) sublines were generated form these original parental radiosensitive (RS) cell lines. Gene expression profiles of radiosensitive (RS) and radioresistant (RR) prostate cancer cell lines were measured.

DNA methyltransferase inhibitor 5-aza-dC-treated prostate cancer cell lines

(Submitter supplied) This microarray dataset accompanies the publication by Kim et al, "The retinoic acid synthesis gene ALDH1a2 is a candidate tumor suppressor in prostate cancer". The dataset comprises four prostate cancer cell lines treated with the DNA methyltransferase inhibitor 5-aza-dC. For each cell line, gene expression in treated (channel 2; Cy5) and untreated (channel 1; Cy3) cells is directly compared by hybridization to the same DNA microarray. more...

DNA methyltransferase inhibitor 5-aza-2'-deoxycytidine effect on prostate cancer cell lines

Analysis of prostate cancer cell lines after treatment with the DNA methyltransferase inhibitor 5-Aza-2'-deoxycytidine (5-aza-dC). Aberrant DNA methylation is an early event in the development of prostate cancer. Results identify hypermethylated genes whose expression is induced by 5-aza-dC.

RNASeq analysis of human ALDH- and ALDH+ DU145 xenografts in nu/nu mice upon radiotherapy treatment

(Submitter supplied) Purpose: we aimed at identify and compare the transcriptional changes of ALDH- and ALDH+ DU145 xenografts upon radiotherapy treatment. Method: Xenografts were generated by injection of ALDH- and ALDH+ DU145 cells in male NMRI-Foxn1 nu/nu immune-deficient mice and subjected to fractionated irradiation to a final dose of 50 Gy. Tumors were excised and processed for total RNA extraction and RNAseq analysis.

Evaluation of gene expression between ALDH-high and ALDH-low cells in endometrial cancer-derived spheroids

(Submitter supplied) To identify genes that are differentially expressed between ALDH-high and ALDH-low cells in endometrial cancer-derived spheroids, we performed whole genome microarray expression profiling as a discovery platform. Human endometrial cancer stem spheroid cells from three patients were sorted into ALDH-high and ALDH-low cells using the FACS Aria II Cell Sorter. Total RNA extracted from the cells were labeled with Cy3 and used for microarray analyses with Agilent Whole Human Genome Oligo Microarrays.

Cellular plasticity upon proton irradiation determines tumor cell radiosensitivity [methylation]

(Submitter supplied) Novel particle therapy was implemented into standard-of-care for cancer patients during the last years. However, experimental studies investigating cellular and molecular mechanisms are lacking although prognostic biomarker are urgently needed. The cancer stem cell (CSC)-related marker aldehyde dehydrogenase (ALDH) is known to impact on prostate cancer radiosensitivity through affecting defense against reactive oxygen species (ROS), reducing DNA damage repair and increasing cell survival. more...

WNT and inflammatory signaling distinguish human Fallopian tube epithelial cell populations

(Submitter supplied) Many high-grade serous carcinomas (HGSCs) likely originate in the distal region of the Fallopian tube's epithelium (TE) before metastasizing to the ovary. Unfortunately, molecular mechanisms promoting malignancy in the distal TE are obfuscated, largely due to limited primary human TE gene expression data. Here we report an in depth bioinformatic characterization of 34 primary TE mRNA-seq samples. These samples were prepared from proximal and distal TE regions of 12 normal Fallopian tubes. more...
