Zipper-interacting protein kinase deficiency effect on coronary artery smooth muscle cells in vitro

Analysis of cultured vascular smooth muscle cells following knockdown of zipper-interacting protein kinase (ZIPK). ZIPK is phosphorylated and activated by Rho-associated kinase 1 (ROCK1). These results, together with those from GDS5659, provide further insight into ROCK1 and ZIPK functions.

Effects of knockdown of ROCK1 and ZIPK on gene expression in cultured human vascular smooth muscle cells

(Submitter supplied) Rho-associated kinase (ROCK) and zipper-interacting protein kinase (ZIPK) have been implicated in diverse physiological functions, including smooth muscle contraction, cell proliferation, cell adhesion, apoptosis, cell migration and inflammation. Many aspects of regulation via ROCK and ZIPK, however, remain unclear. In this study, we utilized an siRNA approach to knock down ROCK1 and ZIPK in cultured human arterial smooth muscle cells. more...

Tissue Factor Pathway Inhibitor-2 is Induced by Fluid Shear Stress in Vascular Smooth Muscle

(Submitter supplied) Tissue Factor Pathway Inhibitor-2 is Induced by Fluid Shear Stress in Vascular Smooth Muscle Cells and Affects Cell Proliferation and Survival Introduction: Vascular smooth muscle cells (SMCs) are exposed to fluid shear stress (FSS) after interventional procedures such as balloon-angioplasty. Whereas the effects of hemodynamic forces on endothelial cells are explored in detail, the influence of FSS on smooth muscle cell function is poorly characterized. more...

Human hepatocellular carcinoma MHCC-97H: Control vs ROCK1 overexpressed vs ROCK2 overexpressed

(Submitter supplied) Transcriptional profiling of human hepatoellular carcinoma (HCC) cell line: MHCC-97H comparing control untreated MHCC-97H cells with MHCC-97H cells overexpressed ROCK1 protein. Goal was to determine the effect of ROCK1 on global MHCC-97H cell mRNA and lncRNA expression. Gene expression profiling in three primary human cell lines treated with atorvastatin or an inhibitor of Rho kinase

(Submitter supplied) Inhibitors of Rho kinase (ROCK) are a new class of drugs with potential benefit in oncology, neurology, and fibrotic and cardiovascular diseases. ROCK inhibitors modulate many cellular functions, some of which are similar to the pleiotropic (non-lipid lowering) effects of statins, suggesting additive or synergistic properties. Studies to date have used compounds which inhibit both isoforms of ROCK, ROCK1 and ROCK2. more...

Comparison of Rho kinase inhibitor and atorvastatin effects on primary cell lines

Analysis of three primary cell lines treated with Rho kinase (ROCK) 2 inhibitor SLx-2119 or with atorvastatin. Effects of ROCK inhibitors overlap to some extent with the pleiotropic (nonlipid lowering) effects of statins. Results suggest possible synergistic effects between the two classes of drugs.

Expression of V3 Versican by Arterial Smooth Muscle Cells Promotes Differentiated and Anti-inflammatory Phenotypes

(Submitter supplied) Arterial smooth muscle cells (ASMCs) undergo phenotypic changes during development and pathological processes in vivo and during cell culture in vitro. Our previous studies demonstrated that retrovirally-mediated expression of the versican V3 splice variant (V3) that lacks glycosaminoglycan chains by ASMCs retards cell proliferation and migration in vitro and reduces neointimal thickening, macrophage and lipid accumulation in animal models of vascular injury and atherosclerosis. more...

Role of differentially expressed microRNA-139-5p in the regulation of phenotypic internal anal sphincter smooth muscle tone

(Submitter supplied) The present study focused on establishing the role of microRNA-139-5p (miRNA-139-5p) in the phenotypic expression of basal tone in rat internal anal sphincter (IAS) vs. lack of tone in truly phasic smooth muscle of anococcygeus (ASM), via RhoA-associated kinase (RhoA/ROCK2).

Lnc-31 acts as a molecular scaffold to control the Rock1 mRNA translation

(Submitter supplied) Cytoplasmic long non coding RNAs have been shown to act at many different levels to control post-transcriptional gene expression; though their role in translational control is still poorly understood. Here we show that lnc-31 is a translational activator of Rock1, a negative regulator of myogenesis which prevents the exit of myoblasts from the cell cycle. This activity well correlates with the described role of lnc-31 in supporting myoblast proliferation. more...

RNAi profiling of primary human AML cells identifies ROCK1 as a therapeutic target and nominates Fasudil as an anti-leukemic drug.

(Submitter supplied) Acute myeloid leukemia (AML) is characterized by a marked genetic heterogeneity, which complicates the development of novel therapeutics. The delineation of pathways essential within the patient-individual mutational background might overcome this limitation and facilitate personalized treatment. We report the results of a large-scale lentiviral loss-offunction RNA-interference-(RNAi)-screen in primary leukemic cells. more...

Gene expression profiling of dSur knock-out flies.

(Submitter supplied) In order to investigate the functional relevance of SUR2 (the protein encoded by ABCC9 in humans) for sleep duration, we knocked down the expression of its Drosophila homologue (dSur) in the flies’ nervous system (both central and peripheral). Unlike humans, flies are active predominantly around dawn and dusk and show two large sleep episodes during the day and during the night. Knockdown of dSur dramatically reduced night-sleep, particularly during the first half of the dark period, but had little effect on the flies’ day-sleep. more...

PAIP1 promote the progress of liver cancer via regulating expression of immune and inflammatory response associated genes

(Submitter supplied) Poly(A) binding protein interacting protein 1 (PAIP1) is a translation regulator and also regulate the decay of mRNA. PAIP1 has also been reported to be a marker of increased invasive potential of liver cancer. However, the underlying molecular mechanism of PAIP1 in liver cancer is still unclear. Here, the gene expression profile of HepG2 cells transfected with PAIP1 shRNA (shPAIP1) was compared with cells transfected with empty vector (shCtrl). more...
